宁夏枸杞根腐病病原研究 被引量:10

Studies on the pathogen of root rot of Lycium barbarum in Ningxia

摘要 为了明确宁夏枸杞根腐病的病原种类,以枸杞根腐病患病植株为试材,采用组织分离法进行根腐病病原菌株的分离和纯化,对病原菌进行形态学和分子生物学鉴定,然后依据柯赫氏法则进行病原菌确认。结果表明:从具有枸杞根腐病症状的枸杞根茎部共分离出38株菌株,形态学观察初步鉴定为4类,以LR-1、LR-2、LR-4和LR-5为代表的4类真菌菌株的分离频率依次是44.74%、34.21%、13.16%、7.89%。致病性检测表明这4类均是引起枸杞根腐病的致病真菌。通过rDNA-ITS序列分析表明,宁夏地区枸杞根腐病病原菌为尖孢镰刀菌(Fusarium oxysporum)、立枯丝核菌(Rhizoctonia solani)、木贼镰刀菌(Fusarium equiseti)和变红镰刀菌(Fusarium incarnatum),其中尖孢镰刀菌(F.oxysporum)的分离频率最高,属优势种群;木贼镰刀菌(F.equiseti)和变红镰刀菌(F.incarnatum)可引起枸杞患根腐病为首次报道。 In order to identify the pathogen of L.barbarum root rot in Ningxia, the diseased plants of L.barbarum root rot were used as test materials, the pathogen was isolated and purified by tissue isolation method, and the pathogen was identified by morphology and molecular biology.Then the pathogen was identified according to Koch ’s rule.The results showed that a total of 38 strains were isolated, which were preliminarily identified as 4 types by morphological observation.The isolation frequencies of four kinds of fungal strains represented by LR-1, LR-2, LR-4 and LR-5 were 44.74%,34.21%, 13.16% and 7.89%, respectively.Pathogenicity test showed that these four kinds of fungi were pathogenic fungi causing root rot of L.barbarum.rDNA-ITS sequence analysis showed that the pathogens of L.barbarum root rot in Ningxia were F.oxysporum, R.solani, F.equiseti and F.incarnatum.F.oxysporum had the highest isolation frequency and belonged to the dominant population.F.equiseti and F.incarnatum could cause root rot of L.barbarum for the first time.
