摘要
红色梨备受青睐,具有广阔的市场价值。红梨着色主要取决于花青苷合成与积累,影响花青苷生物合成的因素有很多,其中光照是最主要的因素之一。本实验对苹果梨进行不同的光处理,基于前期实验室的转录组学分析与相关合成梨花青苷合成调控关键基因的克隆分析,进一步挑选Myb4-like1、bHLH130-like、myb-related protein 306、bHLH96-like、Myb4-like2、MYB1R1、bHLH113-like(pyrus_GLEAN_10011886)、WDR、bHLH147、bHLH113-like(pyrus_GLEAN_10005548)、bHLH57-like共11个差异基因在不同的光处理下进行PCR鉴定,结果发现Myb4-like1、bHLH130-like等基因呈现明显上调的趋势,WDR基因差异表达明显且与花青苷含量趋势相同,这可能是由于在光的调控下改变光周期从而改变基因的表达量,进而影响苹果梨花青苷合成与着色。
Red pears are favored by people and have a broad market value. The color of red pear mainly depends on the synthesis and accumulation of anthocyanin, and there are many factors affecting the biosynthesis of anthocyanin,among which light is one of the most important factors. In this study, different light treatments were carried out on the Pingguoli pear. Based on the transcriptomics analysis in the previous laboratory and the cloning analysis of the key genes regulating the synthesis of related synthetic pear anthocyanin, further select Myb4-like1, bHLH130-like,myb-related protein 306, bHLH96-like, Myb4-like2, MYB1 R1, bHLH113-like(pyrus_GLEAN_10011886), WDR,bHLH147, bHLH113-like(pyrus_GLEAN_10005548), bHLH57-like genetic variations in different PCR identification of light treatment, the results found Myb4-like1, bHLH130-like gene significantly increase trends, such as WDR differentially expressed genes and the same as the contents cyanine trend obviously. This may change the photoperiod under the regulation of light, thus changing the expression level of the gene, and thus affecting the synthesis and coloring of Pingguoli pear anthocyanin.
