                    以葛根粉末为原料,使用低共熔溶剂提取葛根黄酮,于250 nm波长测定吸光度,以葛根黄酮得率为指标,在单因素试验的基础上,采用Box-Behnken响应面法优化超声辅助低共熔溶剂提取葛根中葛根黄酮的工艺,考察加热温度、加热时间、超声时间、超声温度对葛根黄酮得率的影响,并以DPPH·自由基和ABTS;自由基清除能力比较了低共熔溶剂提取液和常规溶剂提取液的抗氧化活性。结果表明,在使用含水量为40%、料液比为1:40、摩尔比为1:5的氯化胆碱/乙二醇低共熔溶剂时对葛根黄酮的提取效果最好。最优提取条件为加热温度82.13 ℃、加热时间40.28 min、超声时间30.54 min、超声温度30.39 ℃,此时葛根黄酮得率为15.65 mg/g。葛根黄酮提取液的DPPH·抑制率为86.55%,ABTS;抑制率为98.52%,均高于常规溶剂提取液。该绿色环保的低共熔溶剂能高效提取葛根中的葛根黄酮,且能够保持良好的抗氧化活性。
                
                The extraction process of puerarin from Pueraria lobata root by ultrasonic assisted deep eutectic solvents extraction was optimized with puerarin content as index and P.lobata root powder as raw material.The effects of heating temperature,heating time,ultrasonic time and ultrasonic temperature on puerarin content were investigated.On the basis of single factor,the Box-Behnken response surface was used to optimize the extraction conditions.The antioxidant activities of deep eutectic solvent extract and conventional solvent extract were compared by DPPH· free radical and ABTS;free radical scavenging ability.The results showed that the optimum extraction conditions were as follows:water content of 40%,solid-liquid ratio of 1:40 and molar ratio of 1:5.The optimal extraction conditions were as follows:heating temperature 82.13 ℃,heating time 40.28 min,ultrasonic time 30.54 min,ultrasonic temperature 30.39 ℃,and the yield of puerarin was 15.65 mg/g.The inhibition rate of DPPH· and ABTS;was 86.55% and 98.52%,respectively.The green deep eutectic solvents can effectively extract puerarin from Pueraria lobata root and maintain good antioxidant activity.
