体外模型研究制川乌和白芍配伍中重金属铅和镉毒性

Toxicity analysis of Pb and Cd in the compatibility of Aconitum carmichaeli Debx.and Paeonia lactiflora Pall.by in vitro model

摘要 中草药中重金属含量是重要质量控制指标之一,本文首次采用体外仿生消化-微生物代谢模型对制川乌和白芍进行预处理,模拟制川乌和白芍单煎液及配伍水煎液在人体内消化和微生物代谢过程,研究制川乌和白芍单煎液及不同配伍水煎液中重金属铅(Pb)和镉(Cd)毒性.实验结果表明制川乌药粉中重金属含量Pb为2.88 mg/kg,Cd为0.53 mg/kg,白芍药粉中Pb为2.01 mg/kg,Cd为0.10 mg/kg,均低于国家现行标准(《中华人民共和国药典》2015年版),由此表明我国居民在饮用含制川乌和白芍中草药汤剂时Pb和Cd暴露水平很低.加入仿生消化液和大肠微生物菌液后,仿生消化和代谢食靡中重金属Pb和Cd浓度逐渐稀释而降低,Pb和Cd的形态随着仿生消化和微生物代谢而转化,其毒性也发生相应的变化.本文所建立的体外仿生消化-微生物代谢模型研究重金属毒性,为川乌和白芍生产和使用风险评估提供一定参考. The content of heavy metals in Chinese herbal medicine is one of the important quality control indexes.In this paper,the single decoction of Aconitum carmichaeli Debx.(A)and Paeonia lactiflora Pall.and their different compatibility decoctions were prepared by in vitro bionic digestion and microbial metabolism model for the first time to study the toxicity of Pb and Cd in the compatibility of herbal medicine.Experimental results show that the contents of Pb and Cd in Aconitum carmichaeli Debx.was 2.88 mg/kg and 0.53 mg/kg,respectively;in Paeonia lactiflora Pall.Pb and Cd content was 2.01 mg/kg and 0.10 mg/kg,respectively,According to the limits of heavy metals for herbal medicines in Chinese Pharmacopoeia of 2015 edition,their content were lower than the current national standard,which indicated that the exposure level of Pb and Cd was very low.After adding the bionic digestive juice and large intestinal microorganism fluid,the contents of Pb and Cd gradually diluted in the digestion and metabolism chyme,and the speciation of Pb and Cd transformed with the migration of gastrointestinal digestive juice,then their toxicity was changed.The in vitro bionic digestion and microbial metabolism model method established in this paper for the analysis and evaluation of heavy metal toxicity,it provides reference for the risk assessment of production and use of Aconitum carmichaeli Debx.and Paeonia lactiflora Pall.
