两种常用老年痴呆大鼠模型制备方法对比

摘要 目的通过比较两种不同的老年痴呆大鼠模型,探求更合适的阿尔茨海默病(AD)动物模型制备方法。方法实验设立正常组、模型Ⅰ组和模型Ⅱ组,随机分组,每组各15只大鼠。模型Ⅰ组采用侧脑室注射Aβ25-35,模型Ⅱ组采用D-半乳糖、亚硝酸钠及三氯化铝联合给药,观察两组大鼠造模48 h内和40 d内存活率,并在造模后1周和5周用水迷宫实验观察大鼠学习记忆能力。结果大鼠存活率检测结果显示三组大鼠造模后存活率差异无统计学意义(P>0.05)。水迷宫定位航行实验结果显示,与正常组相比,造模后1周模型Ⅰ组和模型Ⅱ组的逃避潜伏期明显较长(P<0.05);造模后5周模型Ⅰ组与正常组相比差异无统计学意义(P>0.05),而模型Ⅱ组与正常组相比差异有统计学意义(P<0.05)。水迷宫空间探索实验结果显示,比较第一次穿越平台时间和平台所在象限停留时间,造模后1周两模型组与正常组相比差异有统计学意义(P<0.05),且模型Ⅰ组与模型Ⅱ组相比差异有统计学意义(P<0.05);造模后5周模型Ⅰ组与正常组相比差异无统计学意义(P>0.05),模型Ⅱ组与正常组相比差异有统计学意义(P<0.05)。结论两种方法都可制备出痴呆大鼠模型,但模型Ⅰ组更适合用于短期实验而模型Ⅱ组适合用于长期研究。 Objective To explore a more suitable preparation method for the animal models of Alzheimer's disease(AD)by comparing two different rat models of AD.Methods The normal group,the modelⅠgroup and the modelⅡgroup were established and randomly grouped,with 15 rats in each group.The modelⅠgroup received Aβ25-35 by lateral ventricular injection,and the modelⅡgroup received a combined administration of D-galactose,sodium nitrite and aluminum trichloride.T he survival rate of the rats in the two groups was observed within 48 h and 40 d after modeling,and the learning and memory ability of the rats was observed in the water maze experiment at 1 week and 5 weeks after modeling.Results The test results of survival rate of rats showed that there was no significant difference in the survival rate after modeling of the three groups(P>0.05).The results of the water maze localization navigation experiment showed that the escape latency was significantly longer in the modelⅠgroup and the modelⅡgroup than that in the normal group at 1 week after modeling(P<0.05);At 5 weeks after modeling,there was no statistically significant difference between the modelⅠgroup and the normal group(P>0.05),while there was a statistically significant difference between the modelⅡgroup and the normal group(P<0.05).The results of the water maze spatial exploration experiment showed that there were statistically significant differences(P<0.05)between the two model groups and the normal group in terms of the time to cross the platform for the first time and the residence time on the quadrant where the platform was located at 1 week after modeling,and there were statistically significant differences between the modelⅠgroup and the modelⅡgroup(P<0.05);At 5 weeks after modeling,it was found that there were no statistically significant differences between the modelⅠgroup and the normal group in terms of the time to cross the platform for the first time and the residence time on the quadrant where the platform was located(P>0.05),and there were statistically significant differences between the modelⅡgroup and the normal group(P<0.05).Conclusion Both the two methods can be used to prepare rat models of AD,but the modelⅠgroup is more suitable for short-term experiment while the modelⅡgroup is suitable for long-term study.
