miR-34a在NK/T细胞淋巴瘤组织中的表达及生物学特性影响

摘要 目的探究miR-34a在NK/T细胞淋巴瘤组织中的表达及生物学特性影响。方法回顾性分析2018年6月至2021年6月长沙市第三医院血液肿瘤科收治的50例结外鼻型NK/T细胞淋巴瘤患者及25例鼻腔黏膜慢性炎患者,分别作为NK/T组和正常对照组。对正常NK/T细胞淋巴瘤组织进行慢病毒miR-34a高表达(pLenti-miR)及干扰载体(pLenti-anti-miR)构建,分为miR-34a高表达组及miR-34a干扰组,NK/T细胞淋巴瘤细胞作为miR-34a对照组。采用RT-PCR法检测2组患者组织中miR-34a表达水平,MTT法检测细胞生长增殖能力、流式细胞学检测细胞凋亡情况、Transwell小室检测细胞侵袭迁移能力。结果NK/T组中miR-34a相对表达量(4.69±2.52),显著低于正常对照组(8.21±2.17),差异有统计学意义(P<0.05)。重复测量方差分析结果显示,不同组间、不同时间吸光度均存在显著差异(P<0.001),其中24 h时miR-34a干扰组吸光度显著高于miR-34a对照组,48、72 h时miR-34a高表达组吸光度显著低于miR-34a干扰组、miR-34a对照组,72 h时miR-34a干扰组吸光度显著高于miR-34a高表达组、miR-34a对照组,差异均有统计学意义(P<0.05)。miR-34a高表达组凋亡率为(83.24±4.36)%,显著高于miR-34a干扰组、miR-34a对照组[(20.16±0.96)%、(23.14±3.02)%],差异均有统计学意义(P<0.05)。miR-34a高表达组及miR-34a对照组中穿膜细胞数为(27.34±5.20)、(47.22±5.13)个,显著低于miR-34a干扰组[(182.62±9.42)个],差异均有统计学意义(P<0.05)。结论MiR-34a在NK/T细胞淋巴瘤组织中呈低表达,影响生物学特性扮演抑癌基因角色。 Objective To investigate the expression and biological characteristics of miR-34a in NK/T cell lymphoma.Methods A retrospective analysis was conducted in 50 patients with extranodal nasal NK/T cell lymphoma and 25 patients with chronic nasal mucosal inflammation admitted to the Department of Hematology Oncology,the Third Hospital of Changsha City from June 2018 to June 2021,and they were divided into NK/T group and Normal control group.The normal NK/T cell lymphoma tissues were constructed with lentiviral miR-34a high expression(pLenti-miR)and interference vector(pLenti-anti-miR),which were divided into miR-34a high expression group and miR-34a interference group,and NK/T cell lymphoma cells served as miR-34a control group.Then various indexes were detected by various methods,including miR-34a expression levels by RT-PCR,growth and proliferation of cells by MTT,apoptosis by flow cytometry,invasion and migration of cells by Transwell chamber.Results The relative expression of miR-34a in the NK/T group(4.69±2.52)was significantly lower than that in the normal control group(8.21±2.17),and the difference was statistically significant(P<0.05).Repeated ANOVA showed that there were significant differences in absorbance between different groups and at different times(P<0.001),the absorbance of miR-34a interference group was significantly higher than miR-34a control group at 24 h,the absorbance of miR-34a high expression group was significantly lower than the other two groups at 48 h and 72 h,and the absorbance of miR-34a at 72 h in the interference group was significantly higher than the other two groups,and there were significant differences between the three groups(P<0.05).The apoptotic rate of miR-34a high expression group was(83.24±4.36)%,which was significantly higher than that of miR-34a interference group and miR-34a control group[(20.16±0.96)%,(23.14±3.02)%],the differences were statistically significant(P<0.05).The number of transmembrane cells in the miR-34a high expression group and miR-34a control group were(27.34±5.20)and(47.22±5.13),which were significantly lower than those in the miR-34a interference group[(182.62±9.42)],the differences were statistically significant(P<0.05).Conclusion miR-34a is lowly expressed in NK/T cell lymphoma tissues and is a tumor suppressor gene.
