摘要
建立超高效液相色谱-串联质谱(UPLC-MS/MS)法同时测定醋延胡索药材中黄曲霉毒素B1、B2、G1、G2含量的方法。样品以70%甲醇溶液提取,用免疫亲和柱净化,采用多反应检测(MRM)模式进行定量分析。当进样体积为2μL时,黄曲霉毒素B1、B2、G1、G2的质量分别在0.42~20.80、0.14~7.00、0.47~23.69和0.24~11.80 pg范围内与定量离子的色谱峰面积线性关系良好,相关系数分别为0.9994、0.9990、0.9989和0.9985,检出限分别为0.003、0.005、0.018、0.017μg/kg。高、中、低3个加标浓度水平下,4种黄曲霉毒素的平均回收率分别为88.94%、76.82%、104.90%和93.22%,测定结果的相对标准偏差分别为6.61%、3.71%、2.25%和4.18%(n=6)。该方法适用于醋延胡索药材中黄曲霉毒素的同时测定。
A method of simultaneous determination of aflatoxin B1,B2,G1 and G2 in vinegar corydalis by ultra high performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)was established.The samples were extracted with 70%methanol solution,purified with an immunoaffinity column,and analyzed quantitatively by multi reaction detection(MRM)mode.When the injection volume was 2μL,the mass of aflatoxin B1,B2,G1 and G2 had a good linear relationship with the chromatographic peak area of quantitative ions in the range of 0.42-20.80,0.14-7.00,0.47-23.69 and 0.24-11.80 pg,respectively,the correlation coefficients were 0.9994,0.9990,0.9989 and 0.9985,and the detection limits were 0.003,0.005,0.018 and 0.017μg/kg,respectively.The average recoveries of the four aflatoxins were 88.94%,76.82%,104.90%and 93.22%at high,medium and low spiked concentrations,and the relative standard deviations of the determination results were 6.61%,3.71%,2.25%and 4.18%(n=6)for aflatoxin B1,B2,G1 and G2,respectively.The method is suitable for the simultaneous determination of aflatoxin in vinegar corydalis.
