摘要
结合病原菌的形态特征和分子鉴定,明确引起广州市九里香白粉病的病原菌,为有效防治该病害提供理论依据。利用光学显微镜观察病原菌分生孢子的形态学特征,BLAST在线比对病原菌的ITS和LSU rDNA D1/D2区域序列,并分别构建系统发育进化树。BLAST在线分析结果显示,九里香白粉病原菌的ITS序列与多个Erysiphe quercicola菌株的同源性达到99.84%以上,LSU rDNA序列与多个Erysiphe quercicola菌株的同源性均达到100%。结合病原菌形态学特征和ITS、LSU rDNA D1/D2区域序列的进化树分析结果,将引起广州市九里香白粉病的病原菌鉴定为白粉菌属的Erysiphe quercicola。
Pathogens causing powdery mildew of Murraya paniculata in Guangdong are identified based on the morphological characteristics and molecular identification of the pathogens to provide reference for the effective prevention and control of the powdery mildew disease.The morphological characteristics of the conidia of the pathogens were observed by using optical microscope,and the sequences of the rDNA-ITS and LSU rDNA D1/D2 of the pathogens were aligned by using online BLAST and used to construct their phylogenetic trees,respectively.The BLAST online analysis showed that the ITS sequence of the pathogens of the powdery mildew infecting M.paniculata have a homology of more than 99.84%with multiple strains of Erysiphe quercicola,and the homology of the LSU rDNA sequence with that of multiple strains of E.quercicola strains was upto 100%.The pathogens causing powdery mildew of M.paniculata in Guangdong were hence identified as E.quercicola based on the morphological characteristics of the pathogens and the phylogenetic tree analysis of the sequences of ITS and LSU rDNA D1/D2.
