摘要
中性粒细胞是先天免疫系统中关键的细胞之一,参与了机体损伤、感染、炎症等多种病理与生理过程,具有重要的生理与病理作用。近年来的研究表明:中性粒细胞广泛参与了肿瘤的发生、发展、迁移与侵袭,并且不同表型的中性粒细胞在肿瘤中分别表现出促进肿瘤的发生发展与抑制肿瘤的功能,在肿瘤免疫抑制微环境的形成中发挥着重要作用。基于中性粒细胞在肿瘤免疫抑制机制中的重要性,逐渐成为肿瘤治疗潜在的药理靶点与药物递送载体选择,在肿瘤治疗中表现出潜在的应用价值。从中性粒细胞的来源、发育、分化、表型等方面出发,系统地从血管新生、胞外诱捕网、CTC中性粒细胞簇形成、对肿瘤先天免疫、适应性免疫的作用等角度,综述了中性粒细胞在肿瘤发生、发展、迁移、侵袭、免疫抑制中的作用及机制,叙述了中性粒细胞在药物靶点发现与药物载体设计方面的应用和中性粒细胞与肿瘤关系研究中存在的不足,对未来研究方向进行了展望。
Neutrophils are one of the most important cells in the innate immune system,which are well involved in many pathological and physiological processes,such as injury,infection,inflammation and so on.In recent years,studies have shown that neutrophils are widely involved in the occurrence,development,migration,and invasion of tumor,and play an important role in the formation of tumor immunosuppressive microenvironment.Based on the importance of neutrophils in the pathological mechanism of tumor,neutrophils have gradually become a potential pharmacological target cell and a drug delivery carrier for tumor treatment.Herein,based on the origin,differentiation and phenotype of neutrophils in tumor,we systemically reviewed the role and mechanism of neutrophils in tumorigenesis,development,migration,invasion and immunosuppression,in the perspectives of angiogenesis,neutrophil extracellular traps,cell cluster formation,innate immunity and adaptive immunity.In addition,the application of neutrophils in therapeutic targets and drug carrier design was reviewed,and the shortcomings and further study direction in this area was also prospected.
