摘要
目的观察乳腺癌组织中长链非编码核糖核酸Rho鸟苷三磷酸酶激活蛋白5反义RNA(lncRNA ARHGAP5-AS1)的表达变化,并分析患者预后的影响因素。方法选取乳腺癌及癌旁组织各82例份,采用实时荧光定量聚合链反应法(qRT-PCR法)检测上述组织lncRNA ARHGAP5-AS1,分析lncRNA ARHGAP5-AS1表达与乳腺癌患者临床病理参数的关系,比较不同lncRNA ARHGAP5-AS1表达的乳腺癌患者生存率,分析乳腺癌患者的预后影响因素。结果乳腺癌及其癌旁组织lncRNA ARHGAP5-AS1相对表达量分别为4.53±1.32、1.12±0.31,两者比较,P<0.05。lncRNA ARHGAP5-AS1表达与乳腺癌有无淋巴结转移、TNM分期、肿瘤分化程度、Ki-67表达有关(P均<0.05)。lncRNA ARHGAP5-AS1高表达者(lncRNA ARHGAP5-AS1≥4.53,42例)3年OS生存率为57.14%、3年FPS生存率为42.86%,lncRNA ARHGAP5-AS1低表达者(lncRNA ARHGAP5-AS1<4.53,40例)分别为82.50%、67.50%,两者比较,P均<0.05。有淋巴结转移、TNM分期、lncRNA ARHGAP5-AS1高表达是乳腺癌患者不良预后的影响因素(P均<0.05)。结论乳腺癌组织中lncRNA ARHGAP5-AS1表达升高,其过表达与乳腺癌淋巴结转移、分化程度、TNM分期及患者不良预后有关,可作为乳腺癌预后评估的潜在生物学指标,lncRNA ARHGAP5-AS1高表达、淋巴结转移、TNM分期是乳腺癌患者不良预后的影响因素。
Objective To observe the expression changes of long non-coding RNA Rho guanosine triphosphatase activating protein 5 antisense RNA(lncRNA ARHGAP5-AS1) in the breast cancer tissues and to analyze the prognostic factors.Methods Eighty-two cases of breast cancer tissues and 82 cases of paracancerous tissues were selected,and the IncRNA ARHGAP5-AS1 of these tissues was detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR).The relationship between lncRNA ARHGAP5-AS1 expression and clinicopathological parameters of breast cancer patients was analyzed.The survival rate of breast cancer patients with different lncRNA ARHGAP5-AS1 expression was compared,and the prognostic factors for breast cancer patients were analyzed.Results The relative expression levels of lncRNA ARHGAP5-AS1 in the breast cancer and the adjacent tissues were 4.53±1.32 and 1.12±0.31,respective-1 y,with statistically significant difference(P<0.05).The expression of lncRNA ARHGAP5-AS1 was related to lymph node metastasis,TNM stage,degree of tumor differentiation,and Ki-67 expression in the breast cancer(all P<0.05).The 3-year OS and 3-year FPS survival rates were 57.14% and 42.86%,respectively,in patients with high expression of lncRNA ARHGAP5-AS1(lncRNA ARHGAP5-AS1≥ 4.53,42 cases),versus 82.50% and 67.50%,respectively,in patients with low expression of lncRNA ARHGAP5-AS1(lncRNA ARHGAP5-AS1<4.53,40 cases),with statistically significant difference(all P<0.05).Lymph node metastasis,TNM stage,and high expression of lncRNA ARHGAP5-AS1 were the influencing factors for poor prognosis of breast cancer patients(all P<0.05).Conclusions The expression of lncRNA ARHGAP5-AS1 lncreases in the breast cancer tissues,and its overexpression is related to lymph node metastasis,degree of tumor differentiation,TNM stage and poor prognosis of breast cancer.It can be used as a potential biological indicator for prognosis evaluation of breast cancer.The high expression of lncRNA ARHGAP5-AS1,lymph node metastasis,and TNM stage are the influencing factors for poor prognosis of breast cancer patients.
