摘要
[目的]本文旨在研究植物乳杆菌与反刍动物专用复合酶混合处理玉米秸秆的性能和机制,提高玉米秸秆的瘤胃降解率。[方法]利用单因素试验比较植物乳杆菌处理、反刍动物专用复合酶处理及二者混合处理3种方法对玉米秸秆化学成分的影响,综合利用化学分析法、X-射线衍射仪(XRD)和扫描电子显微镜(SEM)分析处理前与处理后玉米秸秆的物理结构和化学成分变化,利用瘤胃降解试验评价玉米秸秆的消化率。[结果]与植物乳杆菌或反刍动物专用复合酶制剂单独处理相比,混合处理对提高玉米秸秆牛瘤胃降解率的效果最佳,其条件为:8×10^(8) CFU·mL^(-1)植物乳杆菌液2 mL、红糖2 g、蒸馏水128 mL、0.01 g·mL^(-1)专用复合酶溶液20 mL、秸秆50 g,混匀,密封在自封袋中于32℃处理168 h。混合处理后,玉米秸秆试样的纤维素质量在纤维素、半纤维素和木质素总质量中的占比提高3.3%,半纤维素质量占比降低7.7%,木质纤维表面产生孔洞和间隙,干物质24 h和48 h消失率分别提高44.7%和14.2%,中性洗涤纤维24 h和48 h消失率分别提高42.1%和16.4%,酸性洗涤纤维24 h和48 h消失率分别提高28.3%和16.7%。[结论]植物乳杆菌与反刍动物专用复合酶混合处理可以显著提高玉米秸秆的瘤胃降解率,为玉米秸秆饲料化利用提供数据支持。
[Objectives]To improve the rumen degradation rate of corn straw,the performance and mechanism of the mixed treatment of corn straw with Lactobacillus plantarum and ruminant complex enzyme were studied.[Methods]The effects of L.plantarum treatment on the chemical composition of corn straw were carried out by single-factor experiment.The effects of three treatment methods,such as the L.plantarum treatment,ruminant complex enzyme treatment,and mixed treatment,were compared.The changes of physical structure and chemical composition of corn straw before and after the treatment were characterized by using the combination methods,such as the chemical analysis,X-ray diffraction(XRD),and scanning electron microscopy(SEM);the digestibility of corn straw was evaluated by using the rumen degradation test.[Results]Compared with the single treatment of L.plantarum or ruminant complex enzyme,the mixed treatment had the best effect on improving the rumen degradation rate of corn straw.The mixed treatment condition was determined as follows:2 mL of 8×10^(8) CFU·mL^(-1) L.plantarum solution,2 g of brown sugar,128 mL of distilled water,20 mL of 0.01 g·mL^(-1) ruminant complex enzyme solution,and 50 g of corn straw were mixed,and the mixture was then treated in a hermetically plastic ziplock bag at 32℃for 168 hours.Compared with the untreated sample,the mass ratio of cellulose of the treated sample in the cellulose,hemicellulose,and lignin increased by 3.3%,while the mass ratio of hemicelluloses of the treated sample decreased by 7.7%;some holes and gaps were generated on the surface of the lignocellulosic fiber of the sample;the disappearance rate of dry matter of treated sample at 24 hours and 48 hours increased by 44.7%and 14.2%,respectively;the disappearance rate of neutral detergent fiber of treated sample at 24 hours and 48 hours increased by 42.1%and 16.4%,respectively;and the disappearance rate of acid detergent fiber of treated sample at 24 hours and 48 hours increased by 28.3%and 16.7%,respectively.[Conclusions]The mixed treatment of L.plantarum and ruminant complex enzyme can significantly improve the rumen degradation rate of corn straw,thereby providing data support for the utilization of corn straw for feed.
