摘要 目的:采用超高效液相色谱-四级杆飞行时间质谱(UPLC-Q-TOF-MSE)技术对玉泉丸的化学成分进行表征。方法:采用Waters ACQUITY UPLC BEH C18色谱柱(2.1 mm×100 mm,1.7μm),以甲醇-水(含体积比0.1%甲酸)为流动相进行梯度洗脱,流速为0.3 mL/min;采用电喷雾离子源(ESI),正、负离子模式分别进行一级、二级质谱数据采集;采用数据库比对和文献报道对玉泉丸的化学成分进行表征。结果:通过一级MS、二级MS/MS高分辨质谱数据,结合数据库和文献报道对玉泉丸中的142个成分进行了初步表征。其中18个成分来源于葛根,17个成分来源于地黄,7个成分来源于天花粉,24个成分来源于麦冬,28个成分来源于五味子,48个成分来源于甘草。结论:该研究采用UPLC-Q-TOF-MSE技术结合数据处理软件MassLynx和UNIFI,对玉泉丸的化学成分进行了表征,为其进一步的药效物质基础研究奠定了基础。 Objective:To characterize the chemical constituents of Yuquan pills by ultra high performance liquid chromatography-quadrupole time-of flight mass spectrometry(UPLC-Q-TOF-MSE).Methods:Waters ACQUITY UPLC BEH C18(2.1 mm×100 mm,1.7μm)column was used to separate components.Methanol-water(including volume ratio of 0.1%formic acid)was used as mobile phase with gradient elution,the flow rate was 0.3 mL/min,primary and secondary mass spectrometric data were collected in the positive and negative ion modes using an electrospray ion source(ESI),the chemical constituents of Yuquan pills were characterized by database comparison and literature report.Results:The primary characterization of 142 components in Yuquan pills was carried out by MS and MS/MS high resolution mass spectrometry data,combined with database and literature reports.Among them,18 components were from Pueraria lobata,17 from Rehmannia glutinosa,7 from Trichosanthis Radix,24 from Ophiopogon japonicus,28 from Schisandra chinensis and 48 from Glycyrrhizae Radix et Rhizoma.Conclusion:In this study,UPLC-Q-TOF-MSE technology is used in combination with data processing software MassLynx and UNIFI to characterize the chemical constituents of Yuquan pills,which lays a foundation for further studies on the basis of pharmacodynamic substance of Yuquan pills.
