摘要
早发性卵巢功能不全(POI)是造成女性卵巢功能下降的疑难疾病,与卵泡颗粒细胞凋亡密切相关。氧感知通路是人体的重要信号通路之一,缺氧诱导因子-1(HIF-1)α作为氧感知通路的核心元件,参与了一系列与POI相关的信号交联机制,并与其他分子和信号通路元件共同介导了颗粒细胞凋亡。过表达的HIF-1α可以上调BNIP3的表达,引发颗粒细胞凋亡;同时,HIF-1α可通过活化的PI3K/Akt/mTOR通路强化颗粒细胞自噬。颗粒细胞自噬和凋亡相互联系,可共同抑制卵母细胞及卵泡的发育,促进卵泡闭锁,引发POI。本文主要从HIF-1α诱导颗粒细胞凋亡和自噬的发生机制及信号通路探讨基于氧感知通路的POI发生机制,旨在为POI发生机制的研究及治疗提供新思路。
Premature ovarian insufficiency(POI)is a tough disease that causes decreased ovarian function in women and is closely related to follicular granulosa cell apoptosis.The oxygen sensing pathway in the human body is one of important signaling pathways.As the core element of the oxygen sensing pathway,hypoxia-inducible factor-1(HIF-1)αparticipates in a series of signal cross-linking related to POI and mediates granulosa cell apoptosis together with other molecules and signaling pathway elements.Overexpressed HIF-1αupregulates the BNIP3 expression,which may induce granulosa cell apoptosis.Meanwhile,HIF-1αcan enhance granulosa cell autophagy through activated PI3K/Akt/mTOR pathway.Autophagy and apoptosis of granulosa cells are interrelated.They can jointly inhibit the development of oocytes and follicles,promote follicular atresia and lead to POI.In this paper,mechanisms and signaling pathways of apoptosis and autophagy induced by HIF-1αin granulosa cells are investigated to explore the pathogenesis of POI based on oxggen sensing pathway,which is aimed to provide a new approach for the research and treatment of POI.
