摘要
假禾谷镰孢菌(Fusariumpseudograminearum)是引起山东省小麦茎基腐病的主要致病菌,严重影响小麦的产量和品质。研究假禾谷镰孢菌的侵染机制,可为小麦茎基腐病的防治提供理论依据。本研究采用PEG介导的原生质体转化法将GFP基因导入假禾谷镰孢菌中获得转化菌株,经PCR和荧光显微镜观察发现该菌株可稳定遗传;致病力测定发现,GFP转化菌株致病力未减弱。转化菌株侵染试验表明,种衣剂包衣处理可有效防止小麦幼苗被假禾谷镰孢菌侵染;相比10、15℃,20℃条件更有利于假禾谷镰孢菌侵入。本研究建立的PEG介导的假禾谷镰孢菌遗传转化体系,可用于该菌致病相关基因功能研究,标记菌株可用于病菌侵入、田间消长动态、侵染循环等病菌发生规律的研究。
Fusarium pseudograminearum is the main pathogen causing wheat crown rot disease in Shandong Province,which seriously threats the production and quality of wheat.The study on infecting mechanisms of F.pseudograminearum can provide some theoretical bases for the prevention and control of wheat crown rot disease.The PEG-mediated protoplast tranformation method was adopted to transfer GFP gene into F.pseudograminearum,and a transformed strain was obtained and verified as stable inheritance through PCR and fluorescence microscope observation.The virulence of this transformed strain was not decreased.The results of infection with this transformed strain to wheat found that seed coating could effectively prevent the infection of F.pseudograminearum.Compared with 10℃ and 15℃,20℃ was more conducive to the invasion of F.pseudograminearum.The PEG-mediated genetic transformation system of F.pseudograminearum was established,which could be used to study the function of pathogenic genes,and the green fluorescent labeled strain could be used to study the rules of pathogen invasion,field growth and loss dynamics and infection cycle.
