摘要
目的近年来越来越多的研究表明,免疫功能激活和继发的炎性细胞因子异常在CFS发病过程中起着重要的作用,但对其下游信号转导途径尚未报道。探究蒙医温针干预慢性疲劳综合征模型大鼠肿瘤坏死因子死亡受体介导的下游信号转导途径及Caspase-3依赖性的细胞凋亡对免疫功能激活和继发性炎症反应的影响。方法将40只SD大鼠随机分为空白组、对照组、模型组、温针组,后两组负重力竭游泳3周造模。蒙医温针干预对照组和温针组顶会穴和心穴,两穴交替隔1天治疗1次,温度40℃,每次15 min,共治疗11次。采用ELISA法检测血清IL-6、TNF-α、DR及Fas的表达水平;Western blot法检测海马Caspase-3蛋白的表达。结果与空白组比模型组血清IL-6、TNF-α、DR及Fas表达明显升高(P<0.01),与模型组比温针组血清IL-6表达明显降低(P<0.01);与模型组比温针组血清TNF-α、DR及Fas表达明显降低(P<0.05);与空白组比模型组海马Caspase-3蛋白表达明显升高(P<0.05),与模型组比温针组海马Caspase-3蛋白表达明显降低(P<0.05)。结论慢性疲劳综合征可能涉及细胞凋亡过度继发炎症反应及免疫功能的紊乱,使机体能量消耗超负荷导致疲劳。蒙医温针对CFS疗效,可能通过调控Fas的接头蛋白FADD与Caspase-8结合及活化,抑制Caspase-3蛋白的激活,减少与DR的结合减轻细胞的凋亡,从而降低IL-6和TNF-α分泌,减少诱导细胞凋亡作用。
Objective In recent years, more and more studies have shown that immune function activation and secondary inflammatory cytokine abnormalities play an important role in the pathogenesis of CFS,but its downstream signal transduction pathway has not been reported.To explore the effects of Mongolian warm acupuncture on the downstream signal transduction pathway mediated by tumor necrosis factor death receptor and Caspase-3 dependent apoptosis in chronic fatigue syndrome model rats on immune function activation and secondary inflammatory response.Methods Forty SD rats were randomly divided into blank group, control group, model group, warm acupuncture group and the latter two groups were exposed to negative gravity swimming for 3 weeks.Mongolian medicine warm acupuncture intervention in the control group and warm acupuncture group selected Dinghu acupoint and Xin acupoint.Two acupoints alternated every 1 d and the temperature was 40 ℃,15 minutes for each time, a total of 11 times.Serum IL-6,TNF-alpha, DR and Fas were detected by ELISA.The expression of Caspase-3 protein in hippocampus was detected by Western blot.Results Compared with those of the blank group, the expressions of serum IL-6,TNF-α,DR and Fas in the model group were significantly increased(P<0.01).Compared with that of the model group, the serum IL-6 expression in the warm acupuncture group was significantly lower(P<0.01).Compared with those of the model group, the expressions of serum TNF-α,DR and Fas in the warm acupuncture group were significantly lower(P<0.05).Compared with that of the blank group, the expression of Caspase-3 protein in the hippocampus of the model group was significantly increased(P<0.05).Compared with that of the model group, the expression of Caspase-3 protein in the hippocampus of the warm acupuncture group was significantly lower(P<0.05).Conclusion Chronic fatigue syndrome may involve excessive inflammatory response and immune function disorder caused by excessive cell apoptosis, resulting in fatigue caused by overloading the body′s energy consumption.Mongolian medicine may inhibit the activation of Caspase-3 protein by regulating the binding and activation of Fas binding protein FADD and Caspase-8,and reduce the binding of DR to reduce cell apoptosis, thereby reducing IL-6 and TNF-α secretion and reducing the induction of apoptosis.
