基于叶绿素荧光参数的不同矾根品种耐热性评价 被引量:4

Heat tolerance evaluation of different varieties of Heuchera spp.based on chlorophyll fluorescence parameters

摘要 【目的】越夏困难甚至死亡是限制许多矾根品种栽培和应用的关键问题。通过对热胁迫处理后不同品种表型和生理变化情况的研究,探讨矾根的耐热性评价方法,能够为其品种特性评价及耐热机理的深入研究奠定良好基础。【方法】本研究采用45℃高温,对37个矾根品种分别开展3和6 h热胁迫处理,并测定相应的热害指数和叶片叶绿素荧光动力学参数。通过对各叶绿素荧光动力学参数之间以及不同时间热胁迫处理后F_(v)/F_(m)值与热害指数的相关性分析,进行矾根品种耐热性分析最佳指标的筛选。【结果】需经过6 h热胁迫处理,不同矾根品种之间的热害指数才会出现明显差异。而供试品种的叶片F_(v)/F_(m)值在热胁迫处理3和6 h后均出现明显下降,且总体呈现随胁迫时间增加而降低的趋势。不同矾根品种耐热性分析的最佳生理指标是经45℃处理3 h后叶片的F_(v)/F_(m)值。【结论】本研究建立了一种能够快速进行矾根品种耐热性评价的方法。根据45℃处理3 h后叶片的F_(v)/F_(m)值,将矾根品种分为3类,即≥0.70(耐热性较强)类型,(0.55,0.70)(耐热性中等)类型和≤0.55(耐热性较弱)类型。据此筛选出‘甜茶’‘橘子酱’等耐热性较强的品种12个,‘桃色火焰’‘莓果’等耐热性中等的品种17个;‘火警’‘柠檬黄’等耐热性较弱的品种8个。本研究能够为夏季适生性强的矾根品种的选育工作提供重要参考。 【Objective】Many varieties of Heuchera spp.had growth difficulties and even death during summer.It was the key problem restricted cultivation and promotion of Heuchera spp..In order to discuss heat tolerance evaluation method of Heuchera spp.,changes of phenotypic and physiological were researched on different varieties.It can provide a solid foundation for the evaluation of variety characteristics and the further study on the heat tolerance mechanism of Heuchera.【Method】37 varieties were treated with 45℃heat stress for 3 and 6 h to measure the corresponding heat injury index and chlorophyll fluorescence kinetics parameters in this study.Through the correlation analysis among various chlorophyll fluorescence kinetics parameters indexes and the correlation analysis between F_(v)/F_(m)value and heat injury index after high temperature stress,the optimal index for heat tolerance evaluation of Heuchera was selected out from them.【Result】The heat injury index between different varieties of Heuchera only had obvious difference after 6 h high temperature stress treatment.All F_(v)/F_(m)values of 37 varieties of Heuchera had significant decrease,and F_(v)/F_(m)values of the most varieties decreased over the time of high temperature stress.According to the analysis,it was considered that the F_(v)/F_(m)value after 45℃heat stress treatment for 3 h was the optimal index for heat tolerance evaluation of Heuchera.【Conclusion】A fast evaluation method of heat tolerance of Heuchera had been built up in this study.Thirty-seven alum root varieties were divided into three types based on F_(v)/F_(m)value after 3 h of heat stress at 45℃.As follows:≥0.70(strong heat tolerance type),(0.55,0.70)(medium heat tolerance type)and≤0.55(weak heat tolerance type).Each type contained 12 varieties including‘Sweet Tea’and‘Marmalade’,17 varieties including‘Peach Flambe’and‘Mei Guo’,and 8 varieties including‘Fire Alarm’and‘Lime Yellow’,respectively.This study can give an important reference for the screening and the breeding of varieties of Heuchera with strong adaptability in summer.
