摘要
目的探究乙型肝炎病毒(HBV)相关慢加急性肝衰竭(ACLF)患者外周血单个核细胞(PBMC)内环状RNA(circRNA)的表达。方法采集2019年1-12月在番禺区中心医院感染科收治的30例HBV相关慢加急性肝衰竭患者和体检中心30例慢性HBV携带者的外周静脉抗凝血6 mL,取5对标本采用高通量测序检测PBMC中circRNA分子的表达,25对标本用荧光定量PCR进行验证,采用SPSS 22.0软件对数据进行统计学分析。结果与慢性HBV携带组比较,在HBV相关慢加急性肝衰竭患者PBMC中共筛选出861个差异表达的circRNAs,其中386个基因表达上调,475个基因表达下调。q RT-PCR验证hsa_circ_0001523与hsa_circ_0003763在25对样本中的表达水平变化均在肝衰竭组高表达于HBV携带组,差异有统计学意义(均P<0.05)。hsa_circ_0001523与hsa_circ_0003763可结合调控多个miRNA表达。结论HBV相关慢加急性肝衰竭患者外周血单个核细胞内差异表达的circRNA可以结合调控miRNA,可能为阐明该病的发病机制提供新的视角。
Objective To investigate the expression of circular RNA(circRNA)in peripheral blood mononuclear cells(PBMC)of patients with hepatitis B virus(HBV)related acute-on-chronic liver failure(ACLF).Methods Thirty patients with HBV-related acute-on-chronic liver failure admitted in Panyu Central Hospital from January 2019 to December 2019 and 30 chronic HBV carriers in physical examination center were enrolled.PBMC were separated.High-throughput sequencing was used to detect the expression of circRNA in PBMC from 5 pairs of samples,and fluorescence quantitative PCR was used to verify 25 pairs of samples.SPSS 22.0 software was used for statistical analysis.Results Compared with chronic HBV carriers,861 circRNA molecules of differential expression were found in patients with HBV-related acute-onchronic liver failure;of which,386 were up-regulated and 475 were down regulated.Two up-regulated circRNAs with large difference multiple and small P value were selected for preliminary verification.The results showed that the up-regulations of hsa-circ-0001523 and hsa-circ-0003763 were consistent with the results of high-throughput sequencing.It was predicted by software that hsa-circ-0001523 and hsa-circ-0003763 could combine and regulate some miRNAs.Conclusion Differential expression of circRNA in PBMC of patients with HBV-ACLF could bind and regulate miRNAs,which might provide a new perspective for elucidating the pathogenesis of HBV-ACLF.
