摘要
目的研究淫羊藿女贞子对自然衰老骨质疏松大鼠骨调节因子的影响。方法 8月龄SPF级雄性SD大鼠饲养至15月龄后,按随机数字表法分为4组:17月龄组、淫羊藿组(2 g/kg)、女贞子组(1.5 g/kg)、淫羊藿女贞子组(配伍组,4∶3,3.5 g/kg),灌胃给予相应的药物治疗2月后处死;另外以6月龄大鼠作为青年对照组,饲养1周后处死。分离右侧股骨,采用免疫荧光法检测7个骨调节因子,包括transforming growth factor-β1(TGF-β1)、bone morphogenetic protein-2 (BMP2)、Wnt5a、osteoprotegerin(OPG)、nuclear factor-kappa B ligand(RANKL)、macrophage colony-stimulating factor(M-CSF)、d2 isoform of vacuolar (H+) ATPase(ATP6V0d2)的蛋白表达。结果与6月龄大鼠比较,17月龄组大鼠骨组织TGF-β1、BMP2、Wnt5a、OPG、RANKL、M-CSF、ATP6V0d2的蛋白表达均显著降低(P<0.01),且OPG/RANKL降低(P<0.01)。与17月龄组老年大鼠比较,淫羊藿、女贞子单用及配伍均能显著上调这7个骨调节因子的蛋白表达(P<0.01或P<0.05),增加OPG/RANK(P<0.05),且配伍组的上述调节作用优于单用组。结论淫羊藿女贞子配伍防治骨质疏松的作用与调节平衡成骨细胞、破骨细胞微环境的骨调节因子的表达,从而调控骨重建稳态有关。
Objective To investigate the effects of the combination of Herba Epimedii and Fructus Ligustri Lucidi on bone regulatory factor in senile osteoporosis(SOP) rats. Methods 8 Month-old SPF male SD rats were randomly divided into four groups: 17 month old group, Herba Epimedii group(HE, 2 g/kg), Fructus Ligustri Lucidi group(FLL, 1.5 g/kg) and the combination of Herba Epimedii and Fructus Ligustri Lucidi group(HE&FLL, 4:3, 3.5 g/kg). The rats were executed after 2 months of treatment;6-month-old rats were used as the young control group. We detected the bone protein expression of OPG, RANKL, M-CSF, ATP6 V0 d2, BMP2, Wnt5 a and TGF-β1 for evaluating bone metabolism. Results HE&FLL stimulated the osteoclastogenesis and osteoblastogenesis by upregulating the protein expression of OPG, RANKL, M-CSF, ATP6 V0 d2, TGF-β1, BMP2 and Wnt5 a;HE&FLL has a better effect on osteoclastogenesis or steoblastogenesis in SOP rats than HE or FLL. Conclusion The effects of HE&FLL on the prevention and treatment of osteoporosis is related to the regulation of the expression of bone regulatory factors in the microenvironment of osteoblasts and osteoclasts, so as to control the steady state of bone remodeling.
