基于网络药理学研究血府逐瘀汤治疗血管性认知功能障碍的作用机制 被引量:7

An exploration in potential mechanism of Xuefu Zhuyu Decoction in treating vascular cognitive impairment based on network pharmacology

摘要 目的基于多成分、多靶点及多通路研究血府逐瘀汤治疗血管性认知功能障碍的作用机制。方法利用中药系统药理学数据库与分析平台(TCMSP)收集血府逐瘀汤的活性成分,应用Swiss Target Prediction反向预测活性成分靶点,应用GeneGard数据库查询血管性认知功能障碍的靶点,借助STRING平台构建蛋白互作网络(PPI),利用Cytoscape 3.6.0软件可视化和筛选出关键靶点。用DAVID平台对关键靶基因进行GO生物过程富集和分析关键靶点的KEGG信号通路富集。结果研究得到88个主要相关活性成分和29个主要潜在靶点,其中有8个关键靶点;富集分析显示关键靶点主要富集于PI3K-Akt信号通路、HIF-1信号通路、MAPK信号通路、TNF信号通路、FoxO信号通路、Ras信号通路等显著相关的44条通路,涉及涉及ERK1和ERK2级联正调控、MAP激酶活性的正调节、MAPK级联、蛋白激酶B信号的正调控等26条生物过程。结论血府逐瘀汤及其主要活性成分主要通过多靶点、多通路发挥治疗糖尿病肾病的作用机制,其中PI3K-Akt信号通路、MAPK信号通路、FoxO信号通路及HIF-1信号通路可能为主要作用途经。 Objective To explore the mechanism of Xuefu Zhuyu Decoction in the treatment of vascular cognitive impairment based on multi-component,multi-target and multi-path study.Methods The active ingredients of Xufu Zhuyu Decoction were collected by using Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).SwissTargetPrediction was used to predict the target of active ingredients,and GeneGard databases were used to query the target of vascular cognitive impairment.The protein interaction network(PPI)was constructed by STRING platform,and the key targets were visualized and screened by Cytoscape3.6.0 software.The DAVID platform was used to enrich the key target genes in GO biological process and to analyze the KEGG signal pathway enrichment of the key targets.Results A total of 88 main active components and 29 potential targets were obtained,including 8 key targets.Enrichment analysis showed that the key targets were mainly enriched in 44 pathways significantly related,such as PI3K-Akt signal pathway,HIF-1 signal pathway,MAPK signal pathway,TNF signal pathway,FoxO signal pathway,Ras signal pathway,etc.,involving the positive regulation of ERK1 and ERK2 cascades and the positive regulation of MAP kinase activity.There were 26 biological processes,such as MAPK cascade and positive regulation of protein kinase B signal.Conclusion Xuefu Zhuyu Decoction and its main active components mainly play a role in the treatment of diabetic nephropathy through multi-target and multi-pathway,in which PI3K-Akt signal pathway,MAPK signal pathway,FoxO signal pathway and HIF-1 signal pathway may be the main pathway.
