摘要
目的:测定生何首乌、清蒸何首乌和黑豆汁制何首乌中大豆苷元、二苯乙烯苷、大黄素及大黄素甲醚的含量,为制定质量控制指标提供理论依据。方法:取黑豆汁制何首乌、清蒸何首乌和生何首乌粉末分别制成供试品溶液,取大豆苷元、黑豆、二苯乙烯苷、大黄素及大黄素甲醚分别制成对照品溶液。采用薄层色谱法对供试品和对照品溶液进行定性分析,采用高效液相色谱法对供试品及对照品溶液进行定量分析,并进行重复性、稳定性、精密度和加样回收率试验。结果:黑豆汁制何首乌溶液色谱中,在与黑豆对照品和大豆苷元对照品相应位置上显示相同颜色荧光斑点,生何首乌、清蒸何首乌溶液色谱中未显示相同颜色荧光斑点。大豆苷元进样量在1.479~18.782μg范围时与对应色谱峰面积积分值存在线性关系。黑豆汁制何首乌的大豆苷元含量高于生何首乌和清蒸何首乌,二苯乙烯苷、大黄素和大黄素甲醚含量低于生何首乌和清蒸何首乌;生何首乌和清蒸何首乌供试品溶液中未检测出大豆苷元。结论:经炮制的何首乌中有效成分含量发生改变,大豆苷元含量可作为黑豆汁制何首乌与其他炮制工艺何首乌的鉴别与质量控制的检测指标。
OBJECTIVE:To measure the contents of isoflavoues aglycone,stibene glucoside,emodin and physcion in raw polygonum multiflorum,steamed polygonum multiflorum and polygonum multiflorum made from black bean juice,so as to provide reference for the formulation of quality control indicators.METHODS:Take polygonum multiflorum made from black bean juice,steamed polygonum multiflorum,and raw polygonum multiflorum powder to make test solution respectively,and take isoflavoues aglycone,black bean,stibene glucoside,emodin and physcion to make reference solution respectively.Thin layer chromatography was adopted to conduct qualitative analysis on test solution and reference solution,and high performance liquid chromatography was adopted to conduct quantitative analysis on test solution and control solution,tests of repeatability,stability,precision and adding sample recovery were preformed.RESULTS:In the chromatography of polygonum multiflorum solution made from black bean juice,fluorescent spots with same color were displayed on the corresponding positions of black bean and isoflavoues aglycone reference substance,while there were no fluorescent spots with same color displayed on the chromatography of raw polygonum multiflorum and steamed polygonum multiflorum.When the sample size of isoflavoues aglycone was in the range of 1.479-18.782μg,it had a linear relationship with the integrated value of corresponding chromatographic peak area.The content of isoflavoues aglycone in polygonum multiflorum made from black bean juice was higher than that in raw polygonum multiflorum and steamed polygonum multiflorum,while the contents of stibene glucoside,emodin and physcion were lower than those in raw polygonum multiflorum and steamed polygonum multiflorum;no isoflavoues aglycone was detected in the test solution of raw polygonum multiflorum and steamed polygonum multiflorum.CONCLUSIONS:Then contents of active ingredient in prepared polygonum multiflorum have changed,the content determination of isoflavoues aglycone can be used as a detection indicator for the identification and quality control of polygonum multiflorum made from black soybean juice and other processing techniques.
