摘要
目的研究分析华佗豆的化学成分,为后续的开发和利用奠定基础。方法采用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱联用法(UPLC-Q-Orbitrap HRMS),以正、负离子模式扫描采集数据,色谱条件为Thermo Gold C18色谱柱(2.1 mm×100 mm,1.9μm),以0.1%甲酸乙腈(A)-1%甲酸水(含10 mmol乙酸铵)(B)为流动相,流量为0.3 mL/min,进行梯度洗脱。通过分析质谱数据,参考相关文献以及谱库比对,对华佗豆的化学成分进行鉴定。结果共鉴定出华佗豆中的15个化合物,其中小檗碱、烟酰胺、咖啡因、柠檬酸、甘露醇、棉籽糖、乳果糖、羽扇豆醇、6,7-dehydro-11-hydroxy-12,13-epoxyterpendole A为首次从该植物中推测鉴定出来。结论建立的方法快速分析鉴别了华佗豆的化学成分,为华佗豆的质量控制提供科学依据。
Objective To study the chemical constituents of Calonyction muricatum(Linn)G.Don in order to lay the foundation for subsequent development and utilization.Methods Ultra-performance liquid chromatographyquadpole-orbitaltrap high resolution mass spectrometry(UPLC-Q-Orbitrap HRMS)are used to collect data in positive and negative ion modes.Chemical constituents of Calonyction muricatum(Linn)G.Don.is performed on an Thermo Gold C18 column(2.1 mm×100 mm,1.9μm),mobile phase is consisted of 0.1%acetonitrile(A)-the 1%formic acid water(containing 10 mmol ammonium acetate)(B)with gradient elution.The flow rate is 0.3 mL/min.The chemical composition of Calonyction muricatum(Linn)G.Don is identified by relative molecular mass and combined with literature data and mass databases comparison.Results A total of 15 compounds in Calonyction muricatum(Linn)G.Don.are identified,including berberine,nicotinamide,caffeine,citric acid,mannitol,cottonseed sugar,lactulose,lupeol,6,7-dehydro-11-hydroxy-12,13-epoxyterpendole A are first identified from this plant.Conclusion The established method can analyze and identify the components rapidly,which provides scientific evidence for study on pharmacodynamics material of Calonyction muricatum(Linn)G.Don.
