摘要
以牡丹籽粕为原料,采用超声波辅助碱溶酸沉工艺,利用氧化还原电位水对其粗蛋白进行提取,在单因素试验基础上,进行正交试验,对料液比、提取时间、提取温度进行了优化,同时将牡丹籽蛋白与大豆蛋白进行比较。结果表明:最佳的工艺条件为料液比1∶25(g/mL)、提取时间50 min、提取温度50℃、pH 9.5,然后以浓盐酸将提取液pH调节为4.0,酸沉30 min,得到牡丹籽粗蛋白,其提取率为86.53%,氨基酸组成合理,具有较高的营养价值。
The crude protein of peony seed meal was extracted by the technology of ultrasonic assisted alkali-solution and acid-isolation with redox potential water.On the basis of single factor experiment,orthogonal experiment was carried out to optimize the ratio of material to liquid,extraction time and extraction temperature.At the same time,peony seed protein was compared with soybean protein.The result showed that the optimum extraction conditions were as follows:the ratio of material to liquid 1∶25(g/mL),extraction time 50 min,extraction temperature 50℃,and pH 9.5;the pH of the extraction solution was adjusted to 4.0 by concentrated hydrochloric acid,and acid precipitation last 30 min.The extraction rate of crude protein was 86.53%,the amino acid composition of the crude protein was reasonable,and it had high nutritional value.
