茵陈蒿汤抑制JAK2/STAT3信号通路调控肝癌细胞增殖与凋亡 被引量:16

Yinchenhao decoction regulate proliferation and apoptosis in hepatocellular carcinoma by JAK2/STAT3 signaling pathway

摘要 目的探索茵陈蒿汤的抗肝癌作用及其机制。方法体外培养HepG2肝癌细胞,分为对照组和茵陈蒿汤低、中、高浓度(12.5、25、50μg/mL)组。通过CCK-8检测细胞活力、EdU染色观察茵陈蒿汤对HepG2肝癌细胞增殖的影响,钙黄绿素/PI细胞染色观察茵陈蒿汤对肝癌细胞存活能力的影响。通过Western blot检测茵陈蒿汤对肝癌细胞内JAK2/STAT3信号通路及其下游细胞增殖和抗凋亡相关靶蛋白的变化。结果CCK-8和EdU染色结果发现与对照组相比,茵陈蒿汤治疗降低了HepG2细胞的细胞增殖能力。钙黄绿素/PI细胞染色结果发现茵陈蒿汤减弱细胞存活能力,诱导肝癌细胞死亡。最后,Western blot结果显示茵陈蒿汤显著抑制HepG2肝癌细胞内JAK2/STAT3信号通路的激活,同时进一步抑制了JAK2/STAT3信号通路下游增殖相关蛋白Cyclin D1、c-Myc以及抗凋亡相关蛋白Mcl-1、Bcl-XL的表达水平。结论茵陈蒿汤通过抑制JAK2/STAT3信号通路抑制肝癌细胞增殖,诱导细胞凋亡。 Objective Yinchenhao decoction is commonly used for liver and gallbladder diseases treatment.However,the mechanism of Yinchenhao decoction in the treatment of hepatocellular carcinoma has not been fully elucidated.Herein,we evaluated the effects of Yinchenhao decoction on HepG2 cells and its anti-hepatoma mechanism.Methods HepG2 cells were cultured in vitro and divided into control group and Yinchenhao decoction low,medium and high concentration(12.5,25,50μg/mL)groups.CCK-8 was used to detect cell viability.EdU staining was used to observe the effect of Yinchenhao Decoction on the proliferation of HepG2 cells.Calcein/PI staining were performed to assess the effects of Yinchenhao decoction on HepG2 apoptosis.Western blot was used to detect the changes of Yinchenhao decoction on the JAK2/STAT3 signaling pathway and its downstream cell proliferation and anti-apoptotic related target proteins in HepG2.Results The results of CCK-8 and EdU staining indicated that treatment with Yinchenhao decoction reduced the cell proliferation ability of HepG2 cells compared with the control group.The results of calcein/PI cell staining showed that Yinchenhao decoction reduced cell viability and induced apoptosis of HepG2 cells.Finally,Western blot results showed that Yinchenhao decoction significantly inhibited the activation of JAK2/STAT3 signaling pathway in HepG2,and further inhibited the expression level of STAT3 downstream proliferation-related proteins Cyclin D1 and c-Myc and anti-apoptosis-related proteins Mcl-1 and Bcl-XL.Conclusion Yinchenhao decoction inhibits the proliferation of hepatocellular carcinoma cells and induces apoptosis by inhibiting the JAK2/STAT3 signaling pathway.
