摘要
采用反相高效液相色谱法,对钩藤不同部位异钩藤碱、钩藤碱、去氢钩藤碱、异去氢钩藤碱4种生物碱类成分进行含量测定。采用Gemini NX C18(250 mm×4.6 mm,5μm)色谱柱,以1%醋酸铵(A)-乙腈(B)为流动相进行梯度洗脱,流速1.0 mL·min^(-1);检测波长为245 nm;柱温25℃;进样量10μL。本方法简便可靠,专属性强,重现性好,适用于钩藤中生物碱的含量测定;钩藤不同部位生物碱类成分含量差异显著。
The contents of isorhynchophylline;rhynchophylline,corynoxeine and isocorynoxeine in different parts of Uncaria were determined by RP-HPLC.The method was performed on a Gemini NX C_(18) chromatographic column(250 mm×4.6 mm,5μm),with 1%ammonium acetate(A)-acetonitrile(B)as the mobile phase with gradient elution,the flow rate was 1.0 mL·min^(-1),the detect wavelength was 245 nm,the column temperature was 25℃,and the injection volume was 10μL.The method was simple,reliable,specific and reproducible.It was suitable for the determination of alkaloids in Uncaria.The contents of alkaloids in different parts of Uncariawere significantly different.
