摘要
目的:建立乌拉尔甘草HPLC指纹图谱,同时测定不同产地乌拉尔甘草中甘草苷和甘草酸的含量,为经典名方物质基准的研究提供参考。方法:采用色谱柱Agilent 5 TC-C18(250 mm×4.6 mm,5μm),以流动相为乙腈-0.05%磷酸溶液梯度洗脱,检测波长为237 nm,流速为1 ml·min-1,柱温为35℃。建立10批乌拉尔甘草指纹图谱,并采用HPLC法测定其甘草苷和甘草酸的含量。结果:指纹图谱相似度均大于0.9,标定20个共有峰。含量测定表明甘草苷和甘草酸含量最高分别为宁夏红寺堡和新疆,而甘肃黑虎村的甘草苷和甘草酸含量均较高。结论:通过乌拉尔甘草HPLC指纹图谱及两个有效成分的含量测定,可用于经典名方物质基准的质量控制。
Objective: To establish an HPLC fingerprint of Glycyrrhiza uralensis Fisch. and to determine the contents of liquiritin and glycyrrhizic acid in Glycyrrhiza uralensis Fisch. from different origins,so as to provide reference for the study of material basis of the classical prescription. Methods: The contents of liquiritin and glycyrrhizic acid were determined on an Agilent 5 TC-C18 column(250 mm×4.6 mm,5 μm) with the mobile phase of acetonitrile-0.05% phosphoricacid in a gradient elution mode at the detection wavelength of 237 nm,the flow rate of 1 ml·min-1 and the column temperature of 35 ℃ . The chromatographic fingerprint evaluation system was used to establish the fingerprints of 10 batches of Glycyrrhiza uralensis Fisch. Results: The fingerprint similarity was greater than 0.9,and 20 common peaks were calibrated. The results showed that the highest contents of liquiritin and glycyrrhizic acid were in products from Hongsibao of Ningxia and Xinjiang,and the contents of liquiritin and glycyrrhizic acid were relatively high in Heihu village of Gansu province. Conclusion: The HPLC fingerprint and the content determination of two active components can be used for the quality control of the classical prescription.
