摘要
目的对比车前与平车前药材的特征图谱及含量差异性。方法利用UPLC建立车前草药材特征图谱,采用Agilent Zorbax SB C18(100 mm×2.1 mm,1.8mm)色谱柱,以乙腈-0.05%磷酸为流动相进行梯度洗脱,流速为0.3 mL/min,柱温30℃,检测波长330 nm,对25批次不同基原车前草药材的特征图谱与4个指标成分进行同时测定,并进行相似度评价、聚类分析、主成分分析。结果建立了车前草药材UPLC特征图谱,其中车前确定7个共有峰、平车前确定7个共有峰,聚类分析可将25个样本按基原聚为2类;通过主成分分析可将不同基原的车前草药材进行区别;大车前苷和木犀草苷均在车前和平车前中被检出,车前草苷D在车前中被检出,毛蕊花糖苷在平车前中被检出。车前中大车前苷含量较平车前高,木犀草苷含量相近。结论本研究建立的方法简便、快捷、可靠,为不同基原车前草药材的质量控制和基原鉴别提供可靠的分析方法。
Objective To compare the characteristic maps and content differences between Plantago asiatica L.and Plantago depressa Willd.Methods The characteristic chromatogram of Plantaginis Herba was established by UPLC.The column was Agilent Zorbax SB C18(100 mm×2.1 mm,1.8 μm) with acetonitrile-0.05% phosphoric formic acid as mobile phase in gradient elution.The flow rate was 0.3 mL/min,the column temperature was 30℃,and the detection wavelength was 330 nm.The UPLC method was used to establish the characteristic chromatograms of 25 batches of Plantaginis Herba and to determine the 4 index components.The data were analyzed by similarity evaluation,cluster analysis and principal component analysis.Results The established UPLC characteristic chromatogram could be used to identify Plantago asiatica L.and Plantago depressa Willd..There were 7 common peaks in Plantago asiatica L.and Plantago depressa Willd.,respectively.Cluster analysis divided 25 samples into 2 categories based on the origins and principal component analysis could also distinguish the two kinds of Plantaginis Herba.Plantagoside and Cynaroside were detected in Plantago asiatica L.and Plantago depressa Willd.,Plantainoside D was detected in Plantago asiatica L.,and Acteoside was detected in Plantago depressa Willd..The content of plantagoside was higher in Plantago asiatica L.than that in Plantago depressa Willd.,and the content of cynaroside was similar.Conclusion The method is simple,efficient and reliable,which can be used for the quality control and identification among different origins of Plantaginis Herba.
