墨旱莲香豆草醚组分提取工艺优化研究

摘要 对墨旱莲中香豆草醚类组分提取工艺进行了优化研究,以得到香豆草醚组分。用正交实验法对提取条件进行了研究,考察了不同条件对墨旱莲提取效率的影响;用HPLC测定提取物中香豆草醚组分含量,以此评价各组实验提取条件的优劣;用聚酰胺柱色谱对提取物进行纯化,摸索并优化聚酰胺柱色谱的纯化条件。结果显示:最佳提取条件为以体积分数为80%的乙醇为溶剂回流提取,料液比1∶15,提取次数3次;对最优醇提工艺进行验证,收率为10.71%,纯度为1.64%;在纯化实验中,丙酮洗脱系统中香豆草醚组分纯度最高,通过聚酰胺柱色谱将提取物中香豆素类成分纯度提高到了7.97%。 In this experiment,the extraction process of coumestans component in Ecliptae Herba was studied to obtain the optimal process.Firstly,the extraction conditions were studied by orthogonal experiment,and the effects of different conditions on the extraction efficiency of Ecliptae Herba were investigated.The contents of the coumestans component in the extractions were determined by HPLC to evaluate the merits of the experimental conditions.Next,the extractions were purified by using polyamide column chromatography to optimize the purification conditions.The results showed that the optimal extraction conditions were as follows:refluxing with 80%ethanol by volume,the ratio of material to liquid was 1∶15,and the number of extractions was 3 times.The alcohol extraction process was verified to reach a yield of 10.71%with 1.64%purity.In the purification experiment,the purity of coumarin coumestans component increased to 7.97%by acetone clution system with polyamide column chromatography.
