摘要
目的检测Survivin和mmp-9在宫颈上皮内瘤样变(CIN)组织及宫颈癌组织中表达,并探讨其临床意义。方法以濮阳市第三人民医院2017年1月至2020年3月收治的86例宫颈癌及宫颈上皮内瘤样变患者为研究对象,分别采用免疫组化(SP法)和Western Blot检测Survivin和mmp-9的表达阳性率及相对表达量,并进一步分析其与患者临床病理特征的差异性和相关性及其对该肿瘤的临床价值与意义。结果与癌旁组织相比,CIN Ⅰ~Ⅲ期及宫颈癌患者组织中Survivin阳性率均显著上升(P均<0.05);与CIN Ⅰ期患者相比,CIN Ⅲ期及宫颈癌患者组织中Survivin阳性率也均显著上升(P均<0.05),而CIN Ⅱ期Survivin阳性率无显著差异(P>0.05)。与癌旁组织相比,CIN Ⅱ、Ⅲ期及宫颈癌患者组织中mmp-9阳性率均显著上升(P均<0.05),而CIN Ⅰ期患者mmp-9阳性率无显著差异(P>0.05);与CIN Ⅰ期患者相比,CIN Ⅲ期及宫颈癌患者组织中mmp-9阳性率也均显著上升(P均<0.05),而CIN Ⅱ期mmp-9阳性率无显著差异(P>0.05)。与癌旁组织相比,CIN Ⅰ~Ⅲ期及宫颈癌患者组织中Survivin和mmp-9相对表达量均显著升高(P均<0.05);与CIN Ⅰ期相比,CIN Ⅱ、Ⅲ期及宫颈癌患者组织中Survivin和mmp-9相对表达量也均显著升高(P均<0.05)。结论Survivin和mmp-9在不同宫颈上皮内瘤样变及宫颈癌中的阳性表达率及表达量均存在显著差异性,可考虑将其表达水平的变化作为宫颈癌病变的诊断依据。
Objective To detect the expression of Survivin and mmp-9 in cervical intraepithelial neoplasia(CIN)tissues and cervical cancer tissues,and investigate their clinical significance.Methods 86 cases of cervical cancer or cervical intraepithelial neoplasia patients were selected as the research objects,and Immunohistochemistry(SP method)and Western Blot were used to detect the positive rate and relative expression of Survivin and mmp-9.And further analyze its difference and correlation with the clinical pathological characteristics of the patient,and its clinical value and significance for the tumor.Results Compared with the adjacent tissues,the positive rate of Survivin in CIN stages Ⅰ-Ⅲ and cervical cancer patients increased significantly(all P<0.05);Compared with CIN stage Ⅰ patients,Survivin positive rates in CIN stage Ⅲ and cervical cancer patients also increased significantly(all P<0.05),while Survivin positive rates in CIN stage Ⅱ were not significantly different(P>0.05).Compared with adjacent tissues,the positive rate of mmp-9 in CIN Ⅱ,Ⅲ and cervical cancer patients was significantly increased(all P<0.05),while the positive rate of mmp-9 in CIN Ⅰ patients was not significantly different(P>0.05);Compared with CIN Ⅰ patients,the positive rate of mmp-9 in CIN Ⅲ and cervical cancer patients also increased significantly(all P<0.05),but there was no significant difference in the positive rate of mmp-9 in CIN Ⅱ(P>0.05).Compared with adjacent tissues,the relative expression levels of Survivin and mmp-9 in CIN stage Ⅰ-Ⅲ and cervical cancer patients were significantly increased(all P<0.05);Compared with CIN Ⅰ,the relative expression levels of Survivin and mmp-9 in CIN Ⅱ,Ⅲ and cervical cancer patients were also significantly increased(all P<0.05).Conclusion There are significant differences in the positive expression rates and expression levels of Survivin and mmp-9 in different cervical intraepithelial neoplasia and cervical cancer.The changes in their expression levels can be considered as the basis for diagnosis of cervical cancer lesions.
