连作龙胆草根际土壤细菌多样性及功能预测分析 被引量:13

摘要 连续种植单一药用植物会导致严重病害,降低产量和质量,而根际微生物与植物健康密切相关。以清原县种植不同年限龙胆草土壤为研究对象,采用16S rRNA基因高通量测序和PICRUSt功能预测相结合的研究方法比较分析不同年限土壤细菌群落结构和潜在功能组成特征。结果表明:4组龙胆草土壤样品通过高通量测序共得到801346条有效序列,可注释到36门51纲112目218科470属。Alpha多样性分析表明,随着种植年限增加,根际土壤细菌丰富度和多样性指数呈逐年下降趋势,种植3年细菌丰度和多样性大幅度降低。主成分分析表明,连作影响根际土壤细菌的群落结构。与对照组相比,变形菌门、放线菌门和酸杆菌门是龙胆草根际土壤的优势菌群。随种植年限增加,优势菌群unidentified_Actinobacteria、Acidobacteriia和unidentified_Gemmatimonadetes芽孢杆菌纲(Bacilli)纲含量显著增加,α-变形菌纲(Alphaproteobacteria)、Δ-变形菌纲(Deltaproteobacteria)和unidentified_Acidobacteria纲含量显著减少。PICRUSt功能预测分析结果表明,土壤细菌主要涉及遗传信息处理、翻译、细胞生长与死亡、折叠、分类和降解、核苷酸代谢等35个子功能。随着单一栽培年限的增加,典型病原体数量增加,有益细菌数量减少,导致根际微生态失衡,从而造成连作障碍。 Replanting of Medicinal plants causes serious diseases and significant decline in yield and quality,and microbes in the rhizosphere are closely linked with plant health.Soil samples were collected from different ages planted Gentians in Qingyuan County.Subsequently,16S rRNA genes high-throughput sequencing and PICRUSt-based functional prediction were performed to detect soil bacterial community structure and potential functional component.A total of 801346 Clean Reads in four groups were obtained,which were classified as 36 phylums,51 classes,112 orders,218 families,470 genera.The results of the analysis of Alpha diversity showed that soil bacterial richness and diversity index showed a downward trend,the soil bacterial abundance and diversity of the three years of planting decreased significantly.The principal component analysis indicated that the community structure of soil bacteria was affected by replanting.Compared with the control group,the dominant flora in the gentian soil was Proteobacteria,Actinomycetes,and Acidobacteria.With the increase of planting years,the content increased significantly was the dominant bacteria group unidentified_Actinobacteria,Acidobacteria and unidentified_Gemmatimonadetes Bacillus;and significantly reduced contents were Alphaproteobacteria,Deltaproteobacteria and Unidentified_Acidobacteria of the delta-proteobacteria.The PICRUSt analysis showed that the functional gene families were mainly related to 35 sub-functions including the genetic information processing,translation,cell growth and death,folding,sorting and degradation,nucleotide metabolism,and so on.Moreover,the population size of typical pathogens increased and the beneficial bacterial population decreased with the increasing years of monoculture,which resulted in the microecological imbalance in Gentians rhizosphere,thereby caused serious replanting diseases in monocropping system.
