摘要
目的研究大蒜素对脑出血大鼠神经功能的作用及机制。方法SD大鼠60只,分为假手术组、模型组、大蒜素低、中、高剂量组(15、30、60 mg/kg)。实验以胶原酶法制备脑出血大鼠模型。腹腔注射给药24 h后,进行大鼠神经功能评分;测定脑组织含水量;HE染色观察脑组织病理变化;免疫印迹分析(Western blot)检测大鼠脑组织神经丝蛋白200(NF200)、微管相关蛋白-2(MAP-2)、髓鞘碱性蛋白(MBP)、生长相关蛋白(GAP43)、腺苷酸活化蛋白激酶α(AMPKα)、糖原合酶激酶-3β(GSK-3β)蛋白表达。结果与假手术组比较,模型组神经功能评分明显升高(P<0.01);与模型组相比,大蒜素低、中、高剂组神经功能评分明显降低(P<0.05或<0.01)。与假手术组比较,模型组脑含水量明显升高(P<0.01);与模型组相比,大蒜素低、中、高剂组脑含水量明显降低(P<0.01)。与假手术组比较,模型组大鼠脑组织NF200、MAP-2、MBP、GAP43蛋白表达下降(P<0.01);与模型组比较,大蒜素低剂组MAP-2、MBP、GAP43表达上升(P<0.05或<0.01),大蒜素中、高剂组NF200、MAP-2、MBP、GAP43蛋白表达上升(P<0.05或<0.01)。与假手术组比较,模型组大鼠脑组织AMPKα、GSK-3β蛋白表达上升(P<0.01);与模型组比较,大蒜素低剂组GSK-3β蛋白表达下降(P<0.05),大蒜素中、高剂组AMPKα、GSK-3β蛋白表达下降(P<0.01)。结论大蒜素能改善脑出血大鼠神经功能,与促进轴突再生相关蛋白表达有关。
Objective To investigate the effects of allicin on neurological function in rats with intracerebral hemorrhage,and to explore its action mechanism.Methods A total of 60 SD rats were randomly divided into sham operation group,model group,and low-dose,medium-dose and high-dose allicin group(15,30,60mg/kg,respectively).The rat models with cerebral hemorrhage were established by collagenase method.At 24h after peritoneal injection medication,the neurological function scoring was performed,and brain tissue water contents were measured.Moreover the pathological changes of brain tissue were observed by means of HE staining,and the expression levels of brain tissue neurofilament protein 200(NF200),microtubule-associated protein-2(MAP-2),myelin basic protein(MBP),growth-associated protein(GAP43),adenylate-activated protein kinaseα(AMPKα)and glycogen synthase kinase-3β(GSK-3β)protein were detected by Western Blot.Results As compared with those in model group,the neurological scores were significantly reduced,and brain water contents were significantly reduced in allicin groups,and HE staining showed that allicin could significantly improve the pathological damage of brain tissue(P<0.05).Western Blot results showed that the expression levels of NF200,MAP-2,MBP,GAP43 protein were increased significantly,but,the expression levels of AMPKα,GSK-3βprotein were significantly decreased in the allicin groups,as compared with those in model group(P<0.05).Conclusion Allicin can improve the neurological function in rats with intracerebral hemorrhage,which is related to promoting axon regeneration-related protein expression.
