液质联用仪分析氨基酸方法的建立及其在枸杞中的应用 被引量:5

Establishment and Application of a New Assay Method for Detection of Amino Acids of Wolfberry Based on Liquid Chromatography-mass Spectrometry

摘要 建立高效准确快速的通用型氨基酸分析方法是食品、农业、医学等科研工作中重要的研究方向。本文基于液相色谱串联三重四极杆质谱方法,建立了测定氨基酸含量的分析方法,并将该方法应用于枸杞中氨基酸含量的检测。试验优化了样品前处理条件和液质联用上机条件,优化条件下17种氨基酸的检出限为0.001~0.013μg/mL,回收率在90.7%~102.5%之间。将该方法用于枸杞中氨基酸含量的测试,检测结果与使用氨基酸分析仪的国标方法检测结果相对误差较小。该方法检测结果可靠,通用性良好,操作简单,灵敏度高,快速高效。使用该方法分析黑果枸杞和红果枸杞中氨基酸总含量平均值分别为10.2%和9.3%,两类果实中氨基酸含量并无显著差异。 Establishing an efficient,accurate,fast and universal amino acid analysis method is important in food,agriculture,and medicine research.In this paper,a new method for determining amino acids was established based on UHPLC-MS/MS,and the method was applied to measure the amino acids in wolfberry.The sample pretreatment technology and test condition were optimized.The experiment results showed that the detection limits of 17 amino acids was 0.001~0.013μg/mL,and the recovery rates was 90.7%~102.5%under the optimized conditions.Then the method was applied in detection of amino acid of wolfberry,which matched well with the results of amino acid analyzer.The results indicate that this method has obvious advantages in simple operation,high sensitivity,high efficiency and great versatility.Using the above method,the average amino acid contents of Lycium ruthenicum Murr and red wolfberry were 10.3%and 9.4%,respectively,showing no significant difference of amino acid contents between the two kinds of wolfberry.
