牦牛ACAA2基因克隆及组织表达分析 被引量:12

摘要 为获得牦牛ACAA2基因CDS区序列,研究该基因在牦牛不同组织中表达情况。利用RT-PCR方法克隆类乌齐牦牛ACAA2基因CDS序列并分析生物信息学,荧光定量PCR(qPCR)方法检测ACAA2基因在麦洼牦牛心脏、肝脏、脾脏、肺脏、肾脏、胸肌和臀肌组织中表达情况。结果表明,ACAA2基因CDS序列全长为1194 bp,开放阅读框为1101 bp(GenBank登录号:MT861180),共编码366个氨基酸;同源性比对分析显示,牦牛和黄牛亲缘关系最近,其次是马,其ACAA2基因核苷酸序列同源性分别达92.0%和88.7%;ACAA2蛋白亲水性平均值为-0.127,不稳定指数(Ⅱ)为40.88,为不稳定蛋白;无信号肽和跨膜结构,属于非分泌蛋白;存在14个丝氨酸(Ser)、10个苏氨酸(Thr)和4个酪氨酸(Tyr)磷酸化位点;组织表达分析显示,ACAA2基因在牦牛肾脏和肝脏组织中表达差异极显著,在胸肌和臀肌中表达量最低。研究为初步探究ACAA2基因在牦牛脂肪代谢中作用提供理论依据。 In order to obtain the CDS region sequence of ACAA2 gene in yak,and to study the expression of this gene in different tissues of yak,experiment used RT-PCR method to clone the ACAA2 gene CDS sequence of Wuqi yak and conduct bioinformatics analysis.Fluorescence quantitative PCR(qPCR)method was used to detect the expression of ACAA2 gene in Maiwa yak heart,liver,spleen,lung,kidney,pectorales and gluteal muscle tissues.The results showed that the CDS sequence of the ACAA2 gene was 1194 bp in length,the open reading frame was 1101 bp(GenBank Accession No.MT861180),and encoded a total of 366 amino acids.The homology comparison analysis showed that the relationship between the yak and the cattle was the closest,followed by horses,and the nucleotide sequence homology of the ACAA2 gene reached 92.0%and88.7%,respectively.The average hydrophilicity of ACAA2 protein was-0.127,and the instability index(II)was 40.88,which was an unstable protein;it had no signal peptide and transmembrane structure,and was a non-secreted protein;there were 14 serine(Ser)10 threonine(Thr)and four tyrosine(Tyr)phosphorylation sites.Tissue expression analysis showed that the expression level of ACAA2 gene was significantly higher in yak kidney and liver tissues,and the expression level was the the lowest in pectoralis and gluteus muscles.This study provided a theoretical basis for preliminary exploration of the role of ACAA2 gene in yak fat metabolism.
