尿源性干细胞外泌体对移动性牙根吸收修复的影响及其作用机制 被引量:5

摘要 目的探讨尿源性干细胞外泌体对移动性牙根吸收修复的影响以及作用机制。方法取10只SD大鼠,建立移动性牙根吸收模型后,随机分为对照组和实验组各5只。实验组大鼠于左侧上颌第一磨牙近中根颊侧黏膜转折处,注射200μL含100μg(蛋白含量)尿源性干细胞外泌体的磷酸缓冲盐溶液,对照组大鼠则在相同位置注射200μL磷酸缓冲盐溶液。两组均每隔3 d注射1次,共注射5次。注射前及注射5次后,采用micro-CT活体成像仪测量大鼠牙根表面吸收陷窝体积和骨小梁相关结构参数[包括骨小梁数目(Tb.N)、骨小梁分离度(Tb.Sp)、骨体积/组织体积(BV/TV)比值、骨小梁厚度(Tb.Th)]。处死大鼠后,取上颌左侧第一磨牙及其周围牙槽骨组织,利用蛋白免疫印迹法测定骨形态发生蛋白2(BMP-2)、骨唾液酸蛋白(BSP)、骨保护素、核因子κB受体活化因子配体(RANKL)蛋白表达水平。结果注射尿源性干细胞外泌体后,实验组大鼠牙根表面吸收陷窝体积小于注射前水平及对照组,BV/TV比值、Tb.Th高于注射前水平及对照组(均P<0.05)。实验组的BMP-2、BSP表达水平均高于对照组(P<0.05),两组的骨保护素、RANKL表达水平差异均无统计学意义(均P>0.05)。结论尿源性干细胞外泌体可促进移动性牙根吸收的修复,有效改善牙槽骨密度,并可上调BMP-2、BSP表达水平上调,从而诱导牙周细胞的增殖分化。 Objective To investigate the effect of urine-derived stem cell exosomes on the restoration of mobile root resorption and its action mechanism.Methods Ten SD rats were selected to develop the model of mobile root resorption,and then were randomly divided into control group(n=5)and experimental group(n=5).Rats in the experimental group received injection of 200μL phosphate buffered saline containing 100μg(protein content)of urine-derived stem cell exosomes in the mesial-buccal mucosa turning of the left maxillary first molar,whereas rats in the control group were injected with 200μL phosphate buffered saline in the same injection site.Both groups underwent one injection every three days for a total of five injections.Before injection and after five injections,the rats were measured with in-vivo micro-CT imaging system for the volume of resorption lacunae at root surface and the relevant structural parameters of trabecular bone(including trabecular number[Tb.N],trabecular separation degree[Tb.Sp],bone volume/tissue volume[BV/TV]ratio,trabecular thickness[Tb.Th]).The rats were sacrificed to collect the tissues of the left maxillary first molar and its surrounding alveolar bone tissues,followed by the determination of the expression levels of bone morphogenetic protein 2(BMP-2),bone sialoprotein(BSP),osteoprotegerin and receptor activator of nuclear factor kappaB ligand(RANKL)protein by using western blot assay.Results After injection of urine-derived stem cell exosomes,rats in the experimental group presented decreased volume of resorption lacunae at root surface,and higher BV/TV ratio and Tb.Th as compared with the pre-injection levels and the control group(all P<0.05).The expression levels of BMP-2 and BSP in the experimental group were higher than those in the control group(P<0.05),there were no statistically significant differences in expression level of osteoprotegerin or RANKL between the two groups(all P>0.05).Conclusion Urine-derived stem cell exosomes can promote the restoration of mobile root resorption,effectively improve alveolar bone density,and up-regulate the expression levels of BMP-2 and BSP,thus inducing the proliferation and differentiation of periodontal cells.
