丹曲胶囊对麻醉大鼠动脉血压、心率、心肌张力时间指数的影响研究 被引量:5

Effects of Danqu Capsule on Arterial Blood Pressure,Heart Rate and Myocardial Tension Time Index in Anesthetized Rats

摘要 背景丹曲胶囊主要用于治疗胸痹心痛病痰瘀毒互结证,但笔者在临床应用中发现其还对心律失常、高血压患者具有一定治疗作用。目的探讨丹曲胶囊对麻醉大鼠血压、心率及心肌张力时间指数的影响。方法2018-08-13至2018-08-31,将50只雄性SD大鼠随机分为水治疗组、小剂量组、中剂量组、大剂量组及卡托普利组,每组10只。在麻醉状态下,五组大鼠分别经十二指肠灌注0.9%氯化钠溶液(1 ml/100 g)、小剂量丹曲胶囊混悬液(0.6 g/kg)、中剂量丹曲胶囊混悬液(1.2 g/kg)、大剂量丹曲胶囊混悬液(2.4 g/kg)、卡托普利(10 mg/kg),采用RM6240 BD型多道生理信号采集处理系统记录各组大鼠给药前及给药后10、20、30、40、50、60、90、120 min血压(包括收缩压、舒张压、平均动脉压、脉压)、心率,并计算心肌张力时间指数。结果小剂量组、中剂量组、大剂量组及卡托普利组大鼠给药后10、20、30、40、50、60、90、120 min收缩压分别低于本组给药前(P<0.05)。卡托普利组大鼠给药后10 min舒张压低于给药前(P<0.05);小剂量组、中剂量组及卡托普利组大鼠给药20 min舒张压分别低于本组给药前(P<0.05);小剂量组、中剂量组、大剂量组及卡托普利组大鼠给药后30、40、50、60、90、120 min舒张压分别低于本组给药前(P<0.05)。大剂量组和卡托普利组大鼠给药后10 min平均动脉压分别低于本组给药前(P<0.05);小剂量组、中剂量组、大剂量组及卡托普利组大鼠给药后20、30、40、50、60、90、120 min平均动脉压分别低于本组给药前(P<0.05)。卡托普利组大鼠给药后10、20、30、40、50、60、90、120 min脉压均低于给药前(P<0.05)。大剂量组大鼠给药后40、50、60、90、120 min心率均低于给药前(P<0.05);卡托普利组大鼠给药后10、20、30、40、50、60、90、120 min心率均低于给药前(P<0.05)。卡托普利组大鼠给药后10 min心肌张力时间指数低于本组给药前(P<0.05);中剂量组大鼠给药后120 min心肌张力时间指数低于给药前(P<0.05);小剂量组、大剂量组及卡托普利组大鼠给药后20、30、40、50、60、90、120 min心肌张力时间指数分别低于本组给药前(P<0.05)。结论小剂量(0.6 g/kg)、中剂量(1.2 g/kg)、大剂量(2.4 g/kg)丹曲胶囊均对麻醉大鼠具有持续降压作用,大剂量丹曲胶囊具有减慢心率的作用,小剂量、大剂量单曲胶囊具有降低心肌张力时间指数的作用,且安全性较高,故认为丹曲胶囊可能对冠心病合并高血压、心率过速患者安全有效,但针对冠心病合并低血压、心率减慢患者应慎用。 Background Danqu capsule is mainly used to treat the syndrome of phlegm,blood stasis and toxin in chest obstruction and heartache,however,the authors also found its the therapeutic role on arrhythmia and hypertension to a certain extent in clinical application.Objective To explore the effects of Danqu capsule on arterial blood pressure,heart rate and myocardial tension time index in anesthetized rats.Methods From August 13th to August 31th in 2018,50 male Sprague-Dawley(SD)rats were randomly divided into five groups with 10 rats in each group,including water treatment group,low-dose group,medium-dose group,large-dose group and captopril group.Under anesthesia,rats in the five groups were perfused with 0.9%sodium chloride solution(1 ml/100 g),low dose Danqu capsule suspension(0.6 g/kg),medium dose Danqu capsule suspension(1.2 g/kg),large dose Danqu capsule suspension(2.4 g/kg)and captopril(10.4 g/kg)respectively.The blood pressure(including systolic blood pressure,diastolic blood pressure,mean arterial pressure,pulse pressure)and heart rate were recorded before administration and after administration at 10,20,30,40,50,60,90,120 min by using RM6240 BD multi-channel physiological signal acquisition and processing system,and the myocardial tension time index were calculated.Results The systolic blood pressure of rats in low-dose group,medium-dose group,large-dose and captopril group at 10,20,30,40,50,60,90,120 min after administration was lower than that before administration,respectively(P<0.05).The diastolic blood pressure in captopril group at 10 min after administration was lower than that before administration(P<0.05);diastolic blood pressure in low-dose group,medium-dose group and captopril group at 20 min after administration was lower than that before administration,respectively(P<0.05);diastolic blood pressure in low-dose group,medium-dose group,large-dose group and captopril group at 30,40,50,60,90,120 min after administration was lower than that before administration,respectively(P<0.05).The mean arterial pressure at 10 min after administration in large-dose group and captopril group was lower than that before administration,respectively(P<0.05);the mean arterial pressure at 20,30,40,50,60,90 and 120 min after administration in low-dose group,medium-dose group,large-dose group and captopril group was lower than that before administration,respectively(P<0.05).The pulse pressure of captopril group at 10,20,30,40,50,60,90,120 min after administration was lower than that before administration(P<0.05).The heart rate of rats in large-dose group at 40,50,60,90 and 120 min after administration was lower than that before administration,respectively(P<0.05);heart rate of rats in captopril group at 10,20,30,40,50,60,90 and 120 min after administration was lower than that before administration(P<0.05).Myocardial tension time index of rats in captopril group at 10 min after administration was lower than that before administration(P<0.05);myocardial tension time index in medium-dose group at 120 min after administration was lower than that before administration(P<0.05);and that in lowdose group,large-dose group and captopril group at 20,30,40,50,60,90 and 120 min after administration was lower than that before administration,respectively(P<0.05).Conclusion Danqu capsule with low dose(0.6 g/kg),medium dose(1.2 g/kg),large dose(2.4 g/kg)all have continuous antihypertensive effect on anesthetized rats,large-dose Danqu capsule attenuates heart rate,both low-dose and large-dose Danqu capsule down-regulate myocardial tension time index with high safety.Therefore,Danqu capsule may be safe and effective for hypertension and tachycardia in patients with coronary heart disease,whereas its application with caution for hypotension and bradycardia in patients with coronary heart disease.
