摘要
目的以长白山地区不同来源龙胆为样品,考察龙胆炮制生产过程中龙胆苦苷和水溶性浸出物含量的变化趋势,研究生产过程中对其品质产生影响的因素。方法应用高效液相色谱技术(HPLC),流动相为甲醇(A):水(B),流速0.8 mL/min,洗脱程序为:0~20 min,20%A,20~80 min,25%~75%A,80~90 min,100%A,90~100 min,20%A,系统检测波长243 nm,柱温35℃,建立龙胆指纹图谱,并测定各批次龙胆药材及各工序加工后所得样品的龙胆苦苷含量,另测定水溶性浸出物含量,评价各工序产龙胆样品品质。结果在波长为243 nm、梯度洗脱的条件下,每批次样品龙胆苦苷含量均符合药典标准,样品质量合格稳定,龙胆苦苷在4.802~28.812 μg(r=1.00)范围内线性良好,加样回收率为99.93%,RSD为1.09%。龙胆指纹图谱中每一批次样品色谱图与对照图谱相似度均大于0.9。水溶性浸出物含量符合《中国药典》标准。龙胆药材经筛选及人工拣选、快速"抢水洗"、闷润2 h、切段后,龙胆苦苷和水溶性浸出物含量变化相对较小,再经自然晾干后,龙胆苦苷含量普遍升高,浸出物含量明显升高。结论龙胆生产过程中干燥工序对龙胆品质影响较大,在实际生产中应特别注意。
Objective To investigate the variation trend of gentiopicroside and water-soluble extract in the production process of gentian from different sources of gentian in Changbai Mountain,and to study the factors affecting its quality during the production process.Methods Applying high performance liquid chromatography (HPLC),the mobile phase is methanol (A):water (B),flow rate 0.8 mL/min,elution procedure:0-20 min,20%A,20-80 min,25%-75%A,80-90 min,100%A,90-100 min,20%A,system detection wavelength 243 nm,column temperature 35℃,establish gentian fingerprint,and measure each batch of gentian medicinal materials and samples obtained after processing.The content of gentiopicroside and the content of water-soluble extractives were measured to evaluate the quality of gentian samples produced in each process.Results Under the conditions of 243 nm wavelength and gradient elution,the content of gentiopicrin in each batch of samples complies with the pharmacopoeia standards,and the sample quality is qualified and stable.Gentiopicroside has good linearity in the range of 4.802-28.812 μg (r=1.00).Sample recovery was 99.93% and RSD was 1.09%.The similarity between the chromatogram of each batch of samples in the gentian fingerprint and the control is greater than 0.9.The content of water-soluble extractives complies with the standards of the Chinese Pharmacopoeia.After the gentian medicinal materials are screened and manually selected,the "gentle wash",stuffy 2 h,and cut into sections,the content of gentiopicroside and water-soluble extractives is relatively small.After being naturally dried,the content of gentiopicroside generally increased,the content of extracts increased significantly.Conclusion The drying process in the production process of gentian has a great influence on the quality,and special attention should be paid in actual production.
