摘要
目的通过网络药理学探讨山药治疗胃溃疡(GU)的作用机制。方法利用TCMSP数据库获取山药的主要化学成分,经Swiss Target Prediction数据库对预测靶点进一步补充。借助DisGeNET、OMIM和Malacard数据库获取GU相关的潜在作用靶点,采用String数据库建立蛋白互作网络(PPI),使用DAVID数据库分析GO功能及KEGG通路,通过Cytoscape V3.7.2软件构建山药化学成分-靶点网络、山药化学成分-GU-靶点网络和PPI网络。结果从山药中筛选出15个化学成分和118个靶点,与GU相关的作用靶点213个,山药治疗GU涉及13个交集靶点。山药治疗GU主要涉及不饱和脂肪酸代谢、细胞增殖调节、免疫系统发育、MAP激酶活性等生物学过程和HIF-1信号通路、VEGF信号通路、TNF信号通路等。结论山药治疗GU涉及多个靶点、生物学过程和信号通路,为其药理机制研究提供新的思路和理论参考。
Objective To explore the mechanism of Chinese Yam in the treatment of gastric ulcer by network pharmacology.Methods The main chemical components of Chinese Yam were extracted by TCMSP database,and the prediction targets were supplemented by the Swiss Target Prediction database.DisGeNET,OMIM and Malacard databases were used to obtain the potential action targets related to gastric ulcer.The PPI was constructed with String database,and the GO function and KEGG pathway were analyzed with DAVID database.The chemical composition-target network,the chemical composition-gastric ulcer-target network and PPI network were constructed by Cytoscape V3.7.2 software.Results 15 chemical constituents and 118 targets were selected from Chinese Yam,213 targets were related to gastric ulcer,and 13 cross targets were involved in treatment of gastric ulcer.The treatment of Chinese Yam on gastric ulcer mainly involved biological processes such as unsaturated fatty acid metabolic process,ccell proliferation,immune system development,MAP kinase activity,HIF-1 signaling pathway,VEGF signaling pathway,TNF signaling pathway and so on.Conclusion The treatment of Chinese Yam on gastric ulcer involves multiple targets,biological process and signaling pathway,which provides new ideas and theoretical references for the study of its pharmacological mechanism.
