摘要
建立了高效液相色谱法测定饲料中甲硝唑(MNZ)、替硝唑(TNZ)、地美硝唑(DMZ)、奥硝唑(ONZ)和异丙硝唑(IPZ)等5种硝基咪唑类药物的方法。样品用乙酸乙酯提取,正己烷-磷酸溶液反萃取,MCX柱净化,采用Agilent Eclipse Plus C18色谱柱(4.6 mm×250 mm,5.0μm)分离五种硝基咪唑类药物,以甲醇和水为流动相进行梯度洗脱,流速0.8 ml/min,采集波长范围为200~400 nm,记录光谱图和320 nm波长处的色谱图。结果显示,五种硝基咪唑类药物的浓度在0.1~25μg/ml范围内的线性良好,平均加标回收率在70.8%~92.1%范围内,RSD(n=5)在5.6%~10.8%之间,检测限均为0.1 mg/kg。本方法快速、准确,可用于饲料中非法添加5种硝基咪唑类药物的定性和定量检测。
A HPLC-DAD method for the determination of metronidazole,tinidazole,dimetridazole,orni⁃dazole and ipronidazole in feed was established.Samples were extracted with ethyl acetate,and puri⁃fied by hexane-phosphoric acid solution.Then the acidic solution was applied to MCX cartridge for further cleanup.The Agilent Eclipse Plus C18 column(4.6 mm×250 mm,5.0μm)was used to sepa⁃rate five nitroimidazals.Gradient elution was conducted with methanol and water as mobile phases at a flow rate of 0.8 ml/min.The scanning range was within 200~400 nm,and the detection wavelength was at 320 nm.The linearity of the five nitroimidazole drugs was good in the range of 0.1~25μg/ml.The average recovery rates were from 70.8%~92.1%,the RSD(n=5)was within the range of 5.6%~10.8%,and the detection limit was all 0.1 mg/kg.This method is rapid and accurate,and can be used for the qualitative and quantitative detection of five kinds of nitroimidazoles in feed.
