摘要
目的:建立高效液相色谱法测定人血浆中塞克硝唑的药物浓度。方法:色谱柱:Thermo ODS-2柱(250 mm ×4.6 mm,5μm),流动相为乙腈-水(20∶80),流速为1.0 mL/min。紫外检测器检测波长为310 nm。结果:人血浆中塞克硝唑线性范围为0.25~100 mg/L,r=0.9996;平均回收率大于90%,低、中、高浓度样品的日内和日间RSD均小于9.00%,各种条件下血浆样品的稳定性良好。结论:本法操作简便快速,灵敏度高,专属性强,适用于塞克硝唑血药浓度测定。
Objective:To establish a HPLC method for the determination of secnidazole concentration in human plasma.Methods:Liquid chromatography was performed on Thermo ODS-2 column(250 mm×4.6 mm,5μm). The mobile phase of a mixture of acetonitrile water(20∶80)was used at a flow rate of 1.0 mL/min. The detection wavelength was at 310 nm.Results:A linearity was obtained within the range of secnidazole concentrations from 0.25 to 100 mg/L(r= 0.999 6). The average recovery was more than 90% and the intra-day and inter-day RSDs were less than 9.00%. The stability of plasma samples under different conditions was good.Conclusion:It was proved that the method is easy for operation,it is sensitive and specific for the determination of the secnidazole concentrations in plasma.
