摘要
目的：建立高效液相色谱法测定人血浆中塞克硝唑的药物浓度。方法：色谱柱：Thermo ODS-2柱（250 mm ×4.6 mm，5μm），流动相为乙腈-水（20∶80），流速为1.0 mL/min。紫外检测器检测波长为310 nm。结果：人血浆中塞克硝唑线性范围为0.25～100 mg/L，r=0.9996；平均回收率大于90%，低、中、高浓度样品的日内和日间RSD均小于9.00%，各种条件下血浆样品的稳定性良好。结论：本法操作简便快速,灵敏度高,专属性强，适用于塞克硝唑血药浓度测定。
Objective：To establish a HPLC method for the determination of secnidazole concentration in human plasma.Methods：Liquid chromatography was performed on Thermo ODS-2 column（250 mm×4.6 mm,5μm）. The mobile phase of a mixture of acetonitrile water（20∶80）was used at a flow rate of 1.0 mL/min. The detection wavelength was at 310 nm.Results：A linearity was obtained within the range of secnidazole concentrations from 0.25 to 100 mg/L（r= 0.999 6）. The average recovery was more than 90% and the intra-day and inter-day RSDs were less than 9.00%. The stability of plasma samples under different conditions was good.Conclusion：It was proved that the method is easy for operation,it is sensitive and specific for the determination of the secnidazole concentrations in plasma.
