摘要
以漳州香水菠萝皮渣为研究对象,采用Box-Behnken双响应面优化法结合Matlab分析法进行多酚提取量与抗氧化活性的双重工艺优化。通过考察乙醇浓度、料液比、提取温度、提取时间四个单因素对菠萝皮渣多酚含量和羟自由基清除率的影响,经双响应面优化得到最优提取工艺为:乙醇浓度80%,料液比1∶45(g/mL),提取温度30℃,提取时间105 min,该工艺条件下得到多酚提取量为(16.89±0.18)mg GAE/g DW,羟自由基清除率为92.52%±0.19%。同时,经Matlab分析,当提取时间取下限值(C=105 min)时,菠萝皮渣多酚提取量(y1)和抗氧化活性(y2)均能够比中间值组(C=120 min)和上限值组(C=135 min)取得更大的理论值;当料液比越趋近1∶45(g/mL),提取温度越接近30℃,y1和y2值越趋近理论最大值。
In this study,Box-behnken dual response surface optimization method combined with Matlab analysis was used to optimize the extract technique especially extraction amount and antioxidant activity of‘Xiangshui’pineapple peel residue polyphenols from Zhangzhou.The influence of four single factors(ethanol concentration,solid-liquid ratio,extraction temperature,extraction time)on the content of pineapple peel residue polyphenols and its hydroxyl radical scavenging rate were investigated.By the dual response surface analysis,the optimal extraction techniques were as follows:ethanol concentration 80%,solid-liquid ratio 1∶45(g/mL),extracting temperature 30℃,extracting time 105 min.Under these conditions,the polyphenol extraction amount was(16.89±0.18)mg GAE/g DW,and the free radical clearance rate was 92.52%±0.19%.Matlab analysis showed that when the extraction time was taken as the lower limit value(C=105 min),both y1 and y2 had larger theoretical values than the median value group(C=120 min)and the maximum value group(C=135 min).And when solid-liquid ratio and extracting temperature were close to 1∶45(g/mL)and 30℃,respectively,y1 and y2 were close to the maximum theoretical value.
