摘要
目的分析分离纯化的猪肚菇多糖(panus giganteus polysaccharide,PGP)组成及其结构表征,探究PGP对乳腺肿瘤的作用机制。方法采用HPLC分析PGP的纯度和相对分子质量;红外光谱检测糖苷键构型;气相色谱分析糖苷键类型;利用HE染色、TUNEL染色以及免疫组化分析病理切片;Western blot检测与肿瘤细胞增殖、凋亡和转移等相关蛋白的表达水平。结果PGP的相对分子质量为871.331 ku,纯度可达100%,为单组份多糖;红外光谱结果显示PGP为β-糖苷键构型;气相色谱结果表明PGP由1-2键,1-3键,1-4键,1-6键构成。动物实验表明PGP抑制乳腺癌小鼠肿瘤生长和肺转移的作用机制:(1)PGP给药组明显下调PARP、PCNA的表达,抑制乳腺肿瘤细胞的增殖;(2)PGP给药组TUNEL染色明显增强,促进凋亡;(3)PGP给药组下调MMP-9、VEGF及α-SMA的表达,抑制乳腺肿瘤细胞肺转移。结论PGP通过抑制乳腺肿瘤细胞增殖,促进凋亡和抑制转移等机制从而控制乳腺肿瘤的发生发展。
Aim To analyze the composition and structural characterization of the isolated and purified Panus giganteus polysaccharide PGP and to explore the mechanism of PGP on breast tumors.Methods The purity and relative molecular weight of PGP were analyzed by HPLC;the configuration of glycoside bond was detected by infrared spectrum;the type of glycoside bond was analyzed by gas chromatography;the pathological section and efficacy were analyzed by HE staining,TUNEL staining and immunohistochemistry;the expression level of proteins related to tumor cell proliferation,apoptosis and metastasis was detected by Western blot.Results The relative molecular weight of PGP was 871.331 ku,and the purity couldreach 100%,which was a single-component polysaccharide.The infrared spectrum results showed that PGP was β-glycoside bond configuration.The gas chromatography results showed that PGP was composed of 1-2 bond,1-3 bond,1-4 bond and 1-6 bond.Animal experiments showed that PGP could inhibit the growth and lung metastasis of breast cancer mice by the following mechanisms:first,the PGP administration group significantly reduced the expression of PARP and PCNA,and inhibited the proliferation of breast tumor cells;second,TUNEL staining in PGP group was significantly enhanced to promote apoptosis;third,the expression of MMP-9,VEGF and α-SMA in PGP group was down-regulated to inhibit the lung metastasis of breast cancer cells.Conclusions PGP controls the occurrence and development of breast tumors by inhibiting breast tumor cell proliferation,promoting apoptosis and inhibiting metastasis.
