基于网络药理学探讨增液汤对干燥综合征的作用机制 被引量:4

Exploring the mechanism of Zengye Decoction in treating Sjogren’s syndrome based on network pharmacology

摘要 目的基于网络药理学方法探讨增液汤治疗干燥综合征的核心组分及机制。方法采用BATMAN-TCM平台获取增液汤的主要成分及其预测靶点,通过疾病相关的基因与突变位点数据库(DISGeNET)及毒性与基因比较数据库(CTD)获取干燥综合征的靶标基因,并运用Omicshare平台获取增液汤与干燥综合征靶标基因的交集;通过String数据库进行蛋白质相互作用分析,将交集靶点上传到DAVID网站进行GO及KEGG富集分析,应用Cytoscape 3.6.1进行结果分析并将其可视化。结果筛选得到9个增液汤治疗干燥综合征的有效成分,25个核心靶点,DAVID分析得到29个生物过程,25条相关通路。增液汤治疗干燥综合征关键靶点为L-天冬酰胺、甲基麦冬黄烷酮B、二氢麦冬黄酮E,主要作用于PI3K/AKTt通路、哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin, mTOR)通路、Toll样通路等。结论增液汤对干燥综合征的治疗是多成分、多靶点、多通路共同作用的结果,本研究为增液汤治疗干燥综合征作用机制的深入探讨提供了理论依据。 Objective To investigate the core components and mechanism of Zengye Decoction in the treatment of Sjogren’s syndrome (SS) by using network pharmacology. Methods By using BATMAN-TCM database to obtain the main components and targets of Zengye Decoction. The target gene of SS was obtained by DISGeNET database and CTD database. The common part of Zengye Decoction and SS target was obtained by Omicshare platform. Analyzing the common protein interaction by string database and uploading common targets to the DAVID website for GO and KEGG enrichment analysis, then applying Cytoscape 3.6.1 for results analysis and network visualization. Results 9 active components and 25 core targets were screened. DAVID analysis obtained 29 biological processes and 25 related pathways. L-Asparagine, Ophiopogonanone E, Methyl Ophiopogonanone B are the main components acting on PI3K/AKT pathway, mTOR pathway, and TLR pathway, etc. Conclusions Zengye Decoction plays an integrative role of treating Sjogren’s syndrome with the multi-component, multi-target, multi-channel charaters. This paper provides a theoretical basis for further exploration of the mechanism research of Zengye Decoction in the treatment of SS.
