不同宿主动物血清对田鼠型和古典型鼠疫菌抗性的比较研究

摘要 目的研究我国鼠疫菌特性,比较不同宿主动物血清与不同生物型鼠疫菌作用抗性差异。方法无菌条件下分别采集健康成人血清、正常豚鼠血清、正常Balb/c小鼠血清,每种血清分为三组:正常组、灭活组及磷酸盐缓冲液PBS对照组。分别定量感染不同培养温度下的田鼠型和古典型鼠疫菌,比较血清与鼠疫菌作用不同时间的抗性差异及三种不同宿主动物血清间的抗性差异。结果同一种血清与同一生物型鼠疫菌的不同培养温度作用,血清抗性无差异;而同一宿主动物的正常血清与灭活血清、不同宿主动物血清与鼠疫菌的不同作用时间以及不同鼠疫菌之间差异均有统计学意义。结论同一宿主动物血清间比较,灭活血清的抗性效力低于其正常血清;三种宿主动物血清间比较,健康成人血清与小鼠血清对田鼠型鼠疫菌的抗性均强于古典型鼠疫菌,且健康成人血清的抗性效力最强,小鼠血清次之,豚鼠血清差异不明显;宿主血清与不同生物型鼠疫菌的作用时间不同,血清抗性效力也不同。 Objective To study the characteristics of the Yersinia pestis in China,and to compare the difference of resistance to different biotypes of Yersinia pestis among different host animals’serum.Methods The normal human serum(NHS),normal guinea pig serum and normal Balb/c mice serum were collected under aseptic conditions.Each kind of serum was divided into three groups:normal group,inactivation group and PBS control group.Quantitative infections of microtus fuscus and antiqua fuscus Yersinia pestis were carried out at different culture temperatures,respectively.The difference of resistance to Yersinia pestis in serum at different times and the resistance difference in serums of three different host animals were compared.Results There was no difference in serum resistance of the same serum to the same biotype of Yersinia pestis at different culture temperatures.However,there were statistically significant differences between normal serum and inactivated serum of the same host animal,between serum of different host animals and Yersinia pestis at different time,and between serum of different host animals and Yersinia pestis.Conclusion The resistance of inactivated serum was lower than that of normal serum in the same host animal.In the comparison of the serum of three host animals,the resistance of normal human serum(NHS)and Balb/c mice serum to microtus fuscus Yersinia pestis was stronger than that to antiqua fuscus Yersinia pestis.Human serum had the stongest resistance,followed by mice,and the resistance difference of guinea pigs was not obvious.The host animal serum acting on different biotypes of Yersinia pestis at different times will lead to different effectiveness of serum resistance.
