摘要
为了更好地开发利用玉竹,在单因素实验的基础上,利用响应面法对玉竹多糖(PORPs)的提取工艺进行优化,并对PORPs的化学组成、结构及体外抗氧化特性进行研究。获得的PORPs最优提取工艺条件为:提取温度71℃,液料比40.23 mL/g,提取时间1.14 h,此时多糖的提取率达到(5.61±0.12)%。PORPs的化学组成为:总糖含量(76.26±1.53)%,蛋白含量(0.21±0.09)%,糖醛酸含量2.27%。单糖组成为甘露糖(Man):葡萄糖醛酸(GlcA):半乳糖醛酸(GalA):葡萄糖(Glc):阿拉伯糖(Ara):岩藻糖(Fuc)=18.30:0.81:1.46:41.83:1.53:1.06。红外光谱的结果表明,PORPs是一种含有羧基、β型糖苷键、吡喃环结构的多糖。原子力显微镜结果表明,PORPs呈圆顶形颗粒状,直径范围在70~240 nm。扫描电镜图显示,PORPs通过聚集呈现出致密、光滑、带褶皱的片状结构。此外,PORPs展示出了一定的体外抗氧化活性,其中对羟基自由基的清除率达到了(41.98±2.10)%。这项研究对PORPs的开发利用具有理论指导意义。
In order to make full use of Polygonatum ordoratum(Mill.)Druce,the single-factor experiment and response surface experiment were used to optimize the extraction conditions of crude polysaccharide from Polygonatum ordoratum(Mill.)Druce(PORPs).The chemical compositions,structure and antioxidant properties of PORPs were also studied.According to the results,the optimal conditions including extraction temperature,ratio of liquid to solid and extraction time were 71℃,40.23 mL/g and 1.14 h,respectively.And the extraction yield of PORPs was(5.61±0.12)%.The chemical compositions of PORPs including the total sugar content,the total protein content and the content of uronic acid were(76.26±1.53)%,(0.21±0.09)%and 2.27%,respectively.The monosaccharide composition was consisted of mannose,glucuronic acid,galacturonic acid galactose,glucose,arabinose and fucose with a molar ratio of 18.30:0.81:1.46:41.83:1.53:1.06.Furthermore,the FT-IR analysis revealed that POPRs contained-COOH andβ-type glycosidic linkages with pyranose ring.The AFM results suggested that PORPs were dome-shaped particles with particle diameter ranging from 70 nm to 240 nm.In addition,SEM image showed there was a dense,smooth and wrinkled sheet-like structure through aggregation in PORPs.Moreover,PORPs also exhibited free radical scavenging ability in vitro and the scavenging rate of hydroxyl free radical reached(41.98±2.10)%.This study may provide a theoretical guide for the application of PORPs.
