摘要
多糖检测是在生物工程和中药提取与鉴定等研究中最常用的方法之一。利用蒽酮-硫酸法对灵芝多糖进行检测是实验教学中最通用的方法。本文用酶标仪替代传统分光光度计来分析检测多糖,通过对蒽酮-硫酸法在酶标仪上相关使用参数(试剂比例、温度、时间等)优化,建立了用酶标仪分析灵芝多糖的方法,对方法进行了评价。在灵芝多糖分离实验教学中进行了应用实践,取得了满意的结果。该方法能够高通量检测多糖,缩短实验时间,节省大量的危化品试剂,减少废弃试剂处理量。作为科研新方法与传统实验教学相结合的示范,有助于培养学生的创新思维、提高学生分析和解决问题能力。
Polysaccharide detection is one of the most commonly used methods in bioengineering and extraction and identification of traditional Chinese medicine.Anthrone-sulfuric acid method is the most common method for detection of polysaccharide in experimental teaching.In this paper,the traditional spectrophotometer was replaced by the microplate reader to determine the concentration of the polysaccharides.By optimizing the relevant parameters(reagent ratio,temperature,time,etc.)of anthrone-sulfuric acid method when it was used on the microplate reader,a method for the analysis of Ganoderma lucidum polysaccharides by the microplate reader was established,and the method was evaluated.Furthermore,the practical application in the experimental teaching of the separation of Ganoderma lucidum polysaccharide was carried out,and satisfactory results were obtained.It is advantage that it can save experimental time,save a lot of hazardous chemicals reagents,and reduce the amount of waste reagents with this method.At the same time,it will become a demonstration of the combination of new scientific research technology and traditional experimental teaching content,which will help to cultivate students’innovative thinking and improve their ability to analyze and solve problems.
