摘要
为建立高效氯氰菊酯诱导的克氏原螯虾(Procambarus clarkia)肝胰腺损伤模型。分别设置0.005、0.01和0.02μg/L三个浓度高效氯氰菊酯溶液组确定最佳建模浓度,将克氏原螯虾暴露其中,于0.5、1和2 d采样检测相关指标;0.02μg/L高效氯氰菊酯溶液组,在0.5、1、2、3、5、7、14 d分别采集血清检测相关指标确定最适建模天数,并采集肝胰腺进行氧化物、抗氧化物活性检测和病理切片观察。浓度确定试验中,0.02μg/L实验组ALT(谷丙转氨酶)、AST(谷草转氨酶)活性与对照组相比均明显显著升高;天数确定试验中,第3天时实验组ALT、AST活性均显著升高,血清生化指标TP(血清总蛋白)与肝胰腺抗氧化酶SOD(超氧化物歧化酶)、CAT(过氧化氢酶)和氧化产物MDA(丙二醛)与对照相比有显著差异。血清ALP(碱性磷酸酶)和GLU(血糖)未表现出显著差异。病理切片结果显示药物给药3 d后,肝胰腺出现较明显炎症反应。结果表明,0.02μg/L浓度高效氯氰菊酯,给药3 d可以建立克氏原螯虾肝胰腺损伤模型;血清生化指标ALT、AST、TP与肝胰腺抗氧化酶SOD、CAT和氧化产物MDA可作为肝胰腺损伤模型损伤程度判断指标,ALP和GLU不适合作为判断指标。
The Procambarus clarkii was exposed to three concentrations of Beta-cypermethrin at 0.0,0.01 and 0.02μg/L to establish a hepatopancreas model induced by lambda cypermethrin.Serum was collected on 0.5,1 and 2d to detect the activity of transaminase.P.clarkii was exposed to 0.02μg/L cypermethrin solution,and serum was collected at 0.5,1,2,3,5,7 and 14d to detect transaminase activity and pathological observation.Compared with the control group,the activity of aspartate aminotransferase(ALT)and aspartate aminotransferase(AST)in the 0.02μg/L experimental group were significantly increased,the difference was statistically significant.The ALT and AST activities of the experimental group were significantly increased at 3 days.Serum biochemical index total serum protein(TP)was significantly different from hepatopancreas antioxidant enzymes superoxide dismutase(SOD),catalase(CAT)and oxidation products malondialdehyde(MDA)compared with the control gourp.There was no significant difference in alkaline phosphatase(ALP)and blood glucose(GLU).The results of pathological section showed that hepatopancreas had prominent inflammation after three days of drug treatment.The hepatopancreas model of P.clarkii could be established by 0.02μg/L high-efficiency cypermethrintreated for 3 days.The serum biochemical indexes ALT,AST,TP and hepatopancreas antioxidant enzymes SOD,CAT and oxidation product MDA could be used as hepatopancreas injury model Judgment index of damage degree,ALP and GLU were not suitable as judgment indexes.
