家蚕蛹表达对虾白斑综合征病毒囊膜蛋白Vp28和Vp19的免疫原性 被引量:5

Immunogenicity of Prawn White Spot Syndrome Virus Envelope Protein Vp28 and Vp19 Expressed in Silkworm Pupae

摘要 以重组杆状病毒HyNPV-V p28和HyNPV-V p19感染的家蚕蛹研制成白斑综合征病毒(W SSV)囊膜蛋白rV p28和rV p19亚单位疫苗,并对克氏原螯虾持续口服免疫75 d,观察其对W SSV的抗病保护能力。免疫35 d后进行口服攻毒,rV p28、rV p28+rV p19疫苗组的累积死亡率与对照组比较差异显著(P<0.05),免疫保护率(RPS)均达59.26%;注射攻毒后,rV p28疫苗组的累积死亡率比阳性对照组降低40%。第2次口服攻毒,rV p28、rV p28+rV p19疫苗组呈现显著的保护作用,RPS分别达到63.16%、68.42%;第2次注射攻毒,各疫苗组的累积死亡率比对照组降低30%。对免疫组存活虾的胃、肠和肝胰腺组织进行病毒的原位杂交检测均呈阴性反应,而对照组死亡虾的组织都呈阳性反应。结果表明,家蚕蛹表达的病毒囊膜蛋白具有较好的免疫原性,rV p28或者与rV p19合并口服免疫能诱导螯虾产生显著的免疫保护作用。 Vaccines made of envelope proteins Vp28 or Vp19 of white spot syndrome virus (WSSV) expressed in silkworm (13ombyx mori) pupae using recombinant baculovirus were used to investigate the efficacy of oral vaccination on the resistance to WSSV of Procambarus clarkii. It showed that cumulative mortalities after vaccination with rVp28 or rVp28+rVp19 were significantly lower when compared with controls(P〈0.05) following oral challenge on the 35th day post-vaccination(dpv), with relative percent survival(RPS) values of 59.26%. And they also induced a significantly higher resistance to oral re challenge on the 55th dpv(P〈0.05) .with PRS values of 63. 16% and 68.42% ,respectively. A DIG labeled W.SSV DNA probe was used to detect WSSV by in situ hybridization. The positive cells were observed in epithelial cells of stomach,hepatopancreas and gut of the infected control crayfish ,while negative reaction were observed in tissues of survivors-vaccinated. These results indicate Vp28 and Vp19 expressed in silkworm pupae are immunogenic,and immune protection against WSSV can be induced in crayfish by recombinant envelope protein rVp28 and rVp19.
