基于整合药理学平台V2.0的少腹逐瘀汤治疗原发性痛经分子机制研究 被引量:5

摘要 目的:基于整合药理学平台V2.0探讨少腹逐瘀汤治疗原发性痛经的分子机制。方法:通过整合药理学平台V2.0收集少腹逐瘀汤中药成分及其相关靶标、痛经疾病靶标。借助平台分析药物靶标和疾病靶标之间的相互作用关系,构建“中药-疾病核心靶标”网络,最后通过中医药关联网络挖掘模块构建“中药-成分-靶标-通路”多维网络。结果:共收集少腹逐瘀汤9味中药的352种化学成分和1903个药物靶标,筛选获得痛经15个疾病靶标。药物和疾病靶标分析结果显示,共有3个共有靶标(PPARG、PTGS2及CYP17A1);可视化网络分析结果表明,少腹逐瘀汤中的槲皮素、异鼠李素及丁香油酚等成分可能通过作用于ESR1、CNR1及CALM1等靶标参与多巴胺受体、eNOS活化和VEGFA-VEGFR2通路等通路发挥治疗原发性痛经作用。结论:基于整合药理学平台V2.0构建了少腹逐瘀汤与原发性痛经的核心靶标网络和多维可视化网络,为少腹逐瘀汤治疗原发性痛经的研究提供了重要参考,也为临床应用提供了理论依据。 OBJECTIVE:To research the molecular mechanism of Shaofu Zhuyu decoction in the treatment of primary dysmenorrhea based on Integrated Pharmacology Platform V2.0.METHODS:To collect the traditional Chinese medicine components of Shaofu Zhuyu decoction and its related targets and dysmenorrhea disease targets through the Integrated Pharmacology Platform V2.0,so as to analyze the interaction between drug targets and disease targets,to establish a“Chinese medicine-disease core target”network and finally construct a“Chinese medicine-component-target-pathway”multidimensional network through the TCM related network mining module.RESULTS:A total of 352 chemical components and 1903 drug targets of 9 categories of Chinese herbs in Shaofu Zhuyu decoction were collected,15 disease targets of dysmenorrhea were obtained after screening.Results of drug and disease target analysis showed that there were 3 common targets(PPARG,PTGS2 and CYP17A1);results of visual network analysis indicated that quercetin,isorhamnetin and eugenol in Shaofu Zhuyu decoction may participate in pathways of dopamine receptor,eNOS activation and VEGFA-VEGFR2 pathway by acting on targets of ESR1,CNR1 and CALM1,so that play the role in the treatment of primary dysmenorrhea.CONCLUSIONS:The core target network and multidimensional visual network of Shaofu Zhuyu decoction and primary dysmenorrhea are constructed Based on the Integrated Pharmacology Platform V2.0,which provides an important reference for the research of Shaofu Zhuyu decoction in the treatment of primary dysmenorrhea,and also provides a theoretical basis for clinical application.
