摘要
目的研究常春藤皂苷元对乳腺癌细胞MCF-7增殖、凋亡和侵袭的影响及作用机制。方法采用不同浓度常春藤皂苷元处理乳腺癌细胞MCF-7,细胞活力比色分析法(MTS)检测细胞活力,胸腺嘧啶核苷类似物嵌入实验(EdU)检测细胞增殖能力,Hoechst检测细胞凋亡,Western blot检测增殖细胞核抗原(PCNA)蛋白、凋亡相关蛋白(Bax和Bcl-2)和基质金属蛋白酶9(MMP-9)的表达。结果常春藤皂苷元剂量和时间依赖性地抑制MCF-7细胞的生长。MCF-7细胞经常春藤皂苷元干预后,EdU阳性细胞数减少、侵袭细胞数减少和Hoechst阳性细胞数增加。常春藤皂苷元作用MCF-7细胞,可上调Bax的表达和下调Bcl-2、PCNA和MMP-9的表达。结论常春藤皂苷元可抑制乳腺癌细胞MCF-7的增殖、诱导凋亡和减少侵袭,其机制可能分别涉及调节PCNA、Bcl-2/Bax和MMP-9的表达。
Objective To investigate the anti-breast cancer effect of Hederagenin and its mechanism in vitro.Methods MCF-7 cells were treated with different concentrations of ivy saponins.Cell viability was detected by Cell Proliferation Colorimetric Assay Kit(MTS),proliferation was detected by 5-Ethynyl-2’-deoxyuridine(EdU).Hoechst were used to measure apoptosis.The protein expression including proliferating cell nuclear antigen(PCNA),apoptosis-related proteins(Bax and Bcl-2)and matrix metalloproteinase 9(MMP-9)were evaluated by Western blot.Results Hederagenin inhibited the cells viability of MCF-7 in a dose-and time-dependent manner.The EdU positive cells decreased,Hoechst positive cells increased and invasive cells decreased after treating with Hederagenin in MCF-7.Moreover,Hederagenin up-regulated Bax expression and down-regulated Bcl-2,PCNA as well as MMP-9 expression.Conclusion Hederagenin could inhibit the MCF-7 cells proliferation,promote cells apoptosis and decrease cells invasion,which may involve the regulative effect on PCNA,Bax/Bcl-2 and MMP-9 expression.
