碳源和细胞分裂素对‘库尔勒香梨’离体叶片再生不定芽的影响 被引量:2

Effects of Carbon Sources and Cytokinin on Adventitious Bud Regeneration from Leaf Explants of Pyrus sinkiangensi cv.Korla Fragrant Pear

摘要 以库尔勒香梨无菌苗的离体叶片为外植体,研究了影响库尔勒香梨离体叶片再生不定芽的主要因素。碳源物质不仅影响不定芽的再生率而且影响不定芽的生长表现。不定芽再生率总体表现为以D-山梨醇为碳源的高于以蔗糖为碳源的,但以D-山梨醇为碳源的不定芽玻璃化率明显高于以蔗糖为碳源的。在5 mg/L6-苄氨基腺嘌呤(BA)和30g/LD-山梨醇组合的诱导培养基上,不定芽再生率最高,达96.3%,但不定芽玻璃化率也最高,达60.8%;在BA和蔗糖组合的诱导培养基上,不定芽再生率最高为77.8%,且不定芽玻璃化率低,均在10%以下。综合来看,蔗糖作为碳源更适宜诱导库尔勒香梨离体叶片产生正常健壮不定芽。BA比噻苯隆(TDZ)能显著提高不定芽再生率,且BA诱导的不定芽能够直接伸长生长形成不定梢,而TDZ诱导的不定芽,除1mg/LTDZ和30g/L蔗糖组合外,都需转移到不加TDZ的培养基上才能伸长生长形成不定梢。 In vitro leaves of Pyrus sinkiangensi cv.Korla Fragrant Pear were selected as explants.Major factors affecting adventitious bud formation from in vitro leaves were examined.Carbon sources influenced not only adventitious bud regeneration rate,but also adventitious bud growth behavior.For bud regeneration rate,it generally showed higher under D-sorbitol than sucrose.But the hyperhydricity rate of buds under D-sobitol was significantly higher than that under sucrose.The highest bud regeneration rate was achieved on the induction medium with 5 mg/L 6-benzyl amino adenine(BA)and 30 g/L D-sorbitol,and the regeneration rate was 96.3%,but the hyperhydricity rate of buds was also higher,and reached 60.8%.The regeneration rate was the highest as 77.8%on the induction medium with 6-benzyl amino adenine(BA)and sucrose,and the hyperhydricity rate was lower,which was below 10%.In conclusion,sucrose was more suitable for inducing normal and strong adventitious buds with in vitro leaves of Korla Fragrant Pear as explants.Cytokinin BA significantly improved bud regeneration rate than TDZ,and the adventitious buds could directly elongate to form shoots on the induction medium with BA;however,the adventitious buds couldn’t directly elongate to form shoots on the induction medium with TDZ,exclusive of that at lower concentration of 1 mg/L TDZ combined with 30 g/L sucrose;adventitious buds could elongate to form shoots when adventitious buds were transferred onto the medium without TDZ.
