摘要
目的通过观察TNF-α,TNF-αRⅠ在真皮层的表达水平来证明潞党参口服液对光老化小鼠皮肤组织的抗炎作用。方法将90只小鼠随机分为6组,每组15只,空白对照组、UV模型组、VE组和潞党参口服液低、中、高剂量组,对UV模型组、VE组和潞党参口服液低、中、高剂量组制备光老化模型,空白对照组正常饲养。空白对照组和UV模型组灌胃给予生理盐水,潞党参口服液低、中、高剂量组灌胃给予相应剂量的潞党参口服液,VE组灌胃给予维生素E滴剂,经过UV照射造模,观察小鼠表皮形态学变化,采用免疫组化法检测皮肤组织中TNF-α,TNF-αRⅠ的表达水平。结果空白对照组TNF-α免疫组化阳性表达较低,与空白对照组相比较,UV模型组TNF-α免疫组化阳性表达明显升高(P<0.01),具有统计学意义。与UV模型组相比较,潞党参口服液高、中、低剂量组TNF-α免疫组化阳性表达显著下降(P<0.01),差异具有统计学意义。与潞党参口服液中、低剂量组相比较,潞党参口服液高剂量组TNF-α免疫组化阳性表达明显下降(P<0.01),差异具有统计学意义;与空白对照组相比较,UV模型组TNF-αRⅠ免疫组化阳性表达明显较高(P<0.01),具有统计学意义。与UV模型组相比较,潞党参口服液高、中、低剂量组TNF-αRⅠ免疫组化阳性表达显著下降(P<0.01),差异具有统计学意义。与潞党参口服液中、低剂量组相比较,潞党参口服液高剂量组TNF-αRⅠ免疫组化阳性表达明显下降(P<0.01),差异具有统计学意义。结论通过实验研究证明潞党参口服液对皮肤光老化具有抑制作用。
Objective The anti-inflammatory effect of Ludangshen(Codonopsispilosula in Changzhi)oral liquid on the skin tissues of photoaged mice was demonstrated by observing the expressions of TNF-α and TNF-αRⅠ.Methods Ninety mice were randomly divided into 6 groups,with 15 mice in each group.The UV model group,VE group and the low,medium and high dose groups of Ludangshen(Codonopsispilosula in Changzhi)oral liquid in the blank control group were prepared photoaging modelsand the blank control group were fed normally.The blank control group and the UV model group were given normal saline by gavage.The low,medium and high dose groups of Ludangshen(Codonopsispilosula in Changzhi)oral liquid were given the appropriate doses by gavage,and the VE group were given vitamin E drops by gavage.After UV irradiation,the epidermal morphology of mice was observed,and the expression levels of TNF-α and TNF-αRⅠin skin tissues were detected by immunohistochemical method.Results Compared with the that of the blank control group,the immunohistochemical expression of TNF-α in the UV model group was significantly higher(P<0.01).Compared with that of the UV model group,the immunohistochemical expression of TNF-α in the high dose group was significantly decreased(P<0.01),and the difference was statistically significant.The immunohistochemical expression of TNF-α in the low dose group was also significantly decreased(P<0.01),and the difference was statistically significant.Compared with that of the blank control group,the immunohistochemical expression of TNF-αRⅠin the UV model group was significantly higher(P<0.01),which was statistically significant.Compared with that of the UV model group,the immunohistochemical expression of TNF-αRⅠin the high dose group was significantly decreased(P<0.01),and the difference was statistically significant.The immunohistochemical expression of TNF-αRⅠin the low dose group was also significantly decreased(P<0.01),and the difference was statistically significant.Conclusion it is proved that Ludangshen(Codonopsispilosula in Changzhi)oral liquid has inhibitory effect on skin photoaging.
